This paper reports the in vitro anti-diabetic profile of certain medicinal plants traditionally used in Eritrea for the management of type 2 diabetes. The crude methanolic extracts and fractions of Meriandra dianthera, Aloe camperi, a Polyherb and their fractions were investigated. The in vitro screening of the crude extracts has generally elicited a dose dependent α-glucosidase and α-amylase inhibition activities. M. dianthera displayed the highest α-glucosidase inhibitory activity (IC 50 : 0.074 ± 0.032 mg/mL) at the highest concentration tested (0.800 mg/mL) relative to A. camperi, the Polyherb and Acarbose (IC 50 : 0.37 ± 0.052, 0.56 ± 0.024 and 0.55 ± 0.029 respectively). The α-glucosidase inhibition activities of A. camperi and M. dianthera, except for the Polyherb and Acarbose, were significantly different (P < 0.05) at various ranges of concentrations (0.025 -0.800 mg/mL). The percentage α-amylase inhibitions of M. dianthera, A. camperi, the Polyherb and Acarbose, at the highest concentrations (0.800 mg/mL), were 78.3%, 15.9%, 16.4%, and 82.9% respectively. A. camperi and the Polyherb displayed lower α-amylase inhibitory activities (IC 50 :1.72 ± 0.06 and 2.57 ± 0.07 mg/mL respectively) compared to Acarbose and M. dianthera (IC 50 : 0.31 ± 0.01 and 0.43 ± 0.02 mg/mL respectively). For the α-amylase inhibition activity, even at lower concentrations (0.025 mg/mL), there were statistically significant differences (P < 0.01) among the values generated for all the extracts. The in vitro anti-diabetic screening of the fractions, at 0.800 mg/ml, displayed characteristic enzyme inhibition activities.How to cite this paper: Sium, M., Kareru, P., Kiage-Mokua, B., Sood, K., Langley, J. 534Generally, the non-polar fractions showed higher enzyme inhibitory activities compared to the polar fractions. The essential oils of leaves (from male and female plants), unripe fruits and leaf-buds of different samples of Pistacia atlantica collected from Greek East Aegean islands (Kalimnos and Lesvos) were analysed by GC-MS and GC. Qualitative and quantitative differences among the samples and different organs of the plant were observed. The oils were rich in monoterpenes. The main components in the leaf oil from the female plants were myrcene (17.8%, 24.8%) and terpinen-4-ol (11.6%, 6.0%) in the Kalimnos and Lesvos samples, respectively, while in the leaf oil from the male plants terpinen-4-ol (17.3% Kalimnos) and p-mentha-1(7),8-diene (41.1%, Lesvos) were the dominant constituents. The major components in unripe fruits were terpinen-4-ol (25.7%, 8.9%), myrcene (20.2%, 34.5%) and sabinene (14.9%, 19.5%). In the leaf-buds oils sabinene (52.1%) and α α α α α-pinene (11.6%) were the main constituents in the Kalimnos sample, while in the Lesvos sample the major constituent was p-mentha-1(7),8-diene (42.4%).

ABSTRACT:The aim of present study was to investigate the effects of aerobic training with and without Pistacia-atlantica (Bane) extraction as a plant rich in fatty acids, on rat tissues nesfatin-1/nucleobindin-2 and ghrelin gene expression and also on plasma lipid and lipoproteins profiles.Twenty healthy and intact Wistar rats were obtain and randomly were assigned into saline-control (SC), saline-training (ST), Bane-control (BC), and Bane-training (BT) groups. Training groups ran on a motor driven treadmill (at intensity 25m/min, 5days/wk.) for 8 weeks. Rats were orally received saline or bene solution (100 mg/kg and 7.5ul/g of body weight). Blood was collected to determine biochemical variables and liver and visceral fat were also obtain for Nesfatin-1/nucleobindin-2 and ghrelin mRNA expressions by using a Real-time PCR technique.A higher and significant nesfatin-1 mRNA expressions in liver and visceral fat and liver estradiol were observed in trained groups. However, the level of ghrelin expression was lower in trained groups when compared to control rats. In general, lower tissues nesfatin-1mRNA expression and lower plasma and higher estradiol HDL-C were observed in bane-treated rats. A negative and positive correlation between liver estradiol and liver ghrelin expression, plasma estradiol, HDL-C and visceral fat nesfatin-1 expression were found respectively. The results of present study indicate that exercise combined with bane-treatment induced considerable changes on liver and visceral fat nesfatin-1 and ghrelin expression which was accompanied with a significant change in plasma HDL-C as a cardiovascular risk factor.
