Abstract

ABSTRACT'y-Glutamylamine cyclotransferase, an enzyme that catalyzes the conversion of L-y-glutamylamines to free amines and 5-oxo-L-proline, was found in rab it kidney and in various other tissues. The specificity of this enzyme indicates that it functions in the metabolism of products of transglutaminase action; among its substrates are F4L-'y-yglutamyl}L-lysine a derivative of this peptide in which the a-amino and car xy groups of the lysine moiety are blocked, and L-'y-glutamylpolyamine derivatives.The E-(y-glutamyl)lysine crosslink, formed in proteins by the catalytic action of transglutaminase (reaction 1), -Gin-+ -Lys----

“…Έτσι εξηγείται το γεγονός ότι δεν ανιχνεύονται δεσμοί G-L στα γαλα κτοκομικά προϊόντα, αφοΰ το ίδιο το γάλα δεν περιέ χει ενδογενή TG. Πρόσφατα, μάλιστα, αποδείχτηκε η ύπαρξη ενός συστατικού στο γάλα βοοειδών, αιγο προβάτων και του ανθρώπου, το οποίο έχει μικρό μο ριακό βάρος και εμποδίζει τη δράση της προστιθέμε νης MTG (De Jong et al 2003).Η άνοδος της θερμοκρασίας στα τρόφιμα, κατά το μαγειρευμά τους, είναι βραδεία και έτσι η ενδογενής TG μπορεί να παραμένει δραστική για χρονικό διά στημα που είναι επαρκές για τη δημιουργία των δε σμών G-L. Επιπρόσθετα, η θερμική επεξεργασία των πρωτεϊνούχων τροφίμων μπορεί να σχηματίζει δεσμούς G-L μεταξύ της γ-καρβοξυλικής ομάδας του γλουταμι-κοΰ οξέος και της ε-αμινοομάδας της λυσινης, μέσω χη μικής αφυδάτωσης.Οι δεσμοί G-L μπορούν να υδρολυθοΰν αποτελε σματικά in vivo από δυο ένζυμα: την κυκλοτρανσφε-ράση της γλουταμυλαμινης, ένα ένζυμο των νεφρών (Fink et al 1980) και τη γ-γλουταμυλοτρανσφεράση, η οποία απαντάται στο κροσσωτό επιθήλιο του εντέρου, στους νεφρούς και στο αίμα (Meister et al 1981 Seguro et al (1996β).
