Galectin-7 Regulates Keratinocyte Proliferation and Differentiation through JNK-miR-203-p63 Signaling

Abstract: Galectin-7, a member of the β-galactoside-binding protein family, is primarily expressed in stratified epithelial cells, including keratinocytes. There is information in the literature suggesting a role for this protein in regulation of keratinocyte survival and growth, but the underlying mechanism remains relatively unknown. Moreover, its expression pattern in the epidermis suggests that it is also involved in the regulation of keratinocyte differentiation. Here, we demonstrate that galectin-7 knockdown resul… Show more

“…Among tumor-related miRNAs, miR-203 is one of the critical miRNAs that plays important roles in regulating proliferation of several types of cells. miR-203 reduced the PDGF-stimulated proliferation in human airway smooth muscle (HASM) cells (27), and formed the JNK-miR-203-p63 signaling to regulate keratinocyte proliferation and differentiation (28). miR-203 also plays important roles in cancerigenesis.…”

“…MicroRNAs play a role in regulating the cashmere growth cycle by targeting various signalling pathways and transcription factors 14 . MiR-203 regulates the differentiation of epithelial keratinocytes and directly inhibits the expression of p63 12 , 15 . MiR-200b and MiR-196a are associated with hair follicle development 16 , 17 .…”

“…Indeed, ectopic expression or addition of exogenous galectin-7 in the DLD-1 human colon carcinoma cell line [ 55 ] and the neuroblastoma cells SK-N-MC, respectively [ 53 ], drastically reduced tumour cell proliferation. Consistently, galectin-7 knockdown in human keratinocytes results in a hyperproliferative phenotype [ 38 ]. However, ectopic expression of galectin-7 in the B16F1 melanoma cell line did not affect cell growth [ 60 ] indicating that cell context might be important for galectin-7 to modulate cell proliferation.…”

“…Regarding tumour biology, galectin-7 downregulation correlates with poor tumour differentiation in bladder squamous cell carcinomas [ 63 ] and in vulvar squamous cell carcinoma [ 64 ]. Recently, Liu and colleagues produced the first mechanistic evidence of a function of galectin-7 in keratinocyte differentiation in vitro [ 38 ]. In fact, in the keratinocyte cell line HaCaT, galectin-7 knockdown reduces cell differentiation as assessed by the expression of keratins.…”
