Gadolinium-Based Hybrid Nanoparticles as a Positive MR Contrast Agent

Abstract: A new nanoparticulate inorganic-organic hybrid-type positive contrast agent (CA), PGP/dextran-K01, was synthesized based on a GdPO4 inorganic core as a relaxation-time-shortening moiety and a dextran-coating, which generates monodispersibility in water, a high relaxation-time-shortening effect by retaining a large number of water molecules in proximity of the core and toxicity reduction in in-vivo studies. This PGP/dextran-K01 nanoparticle has high r1 and r2 values and a significantly low r2/r1 value, 1.1, whi… Show more

“…The r 2 /r 1 of ZES-SPIONs is slightly lower and therefore slightly better than that of VSOP (44), a state-of-the-art SPION-based T 1 contrast agent. The r 2 /r 1 of our ZES-SPIONs is more preferable than that of other SPION-based MRI contrast agents including Feraheme, Resovist, Feridex, Combidex, Supravist, Clariscan, and others (3,8,9,21,46,47). Furthermore, as shown in Fig.…”

“…[46] Dextran-coated GdPO 4 nanoparticles (PGP/dextran-K01) were synthesized by a hydrothermal process in the presence of dextran. [47] These Gd-based nanoparticles had significantly low r 2 /r 1 values, and they showed strong positive-contrast effects. However, uniformly sized nanoparticles of gadolinium or related lanthanide compounds have not yet been demonstrated.…”

“…The most obvious nanoparticles for T1 agents are gadolinium-based ones, because many gadolinium complexes have been extensively used as clinical MRI contrast agents. Nanoparticles of gadolinium oxide (Gd 2 O 3 ), [45] gadolinium fluoride (GdF 3 ), [46] and gadolinium phosphate (GdPO 4 ) [47] have been investigated as MRI contrast agents. They exhibited signal-enhancing contrast in T1-weighted images with low r 2 /r 1 values.…”

“…; Fortin et al 2007;McDonald and Watkin 2006;Park et al 2009;Söderlind et al 2005;Petoral Jr et al 2009;Kim et al 2010), gadolinium fluoride (GdF 3 ) (Evanics et al 2006;Chaput et al 2011) or gadolinium phosphate (GdPO 4 ) (Hifumi et al 2006;Yan et al 2010;Lai et al 2008). The crystals preferably are kept small enough to give a large surface to bulk ratio and thereby an efficient interaction between Gd 3+ and water molecules; i.e a high water exchange.…”
