“…Nonetheless, it remains possible that the ICV α-MSH treatment reduced the passive responses by some means other than its anti-inflammatory activity. α-MSH is an endogenous peptide that plays a role in appetite, anxiety, and other motivated responses (Gonzalez, Vaziri, & Wilson, 1996;Rao, Kokare, Sarkar, Khisti, Chopde, & Subhedar, 2003). While what is known about these other actions of α-MSH are not easily reconciled with the behavioral effects we observed, we cannot rule out an influence of our ICV infusion of α-MSH on motivational systems independent of inflammatory action.…”

“…Second, the vocalizing of guinea pig pups during isolation procedures is considered to reflect an anxiety-like state (Borsini, Podhorna, & Marazziti, 2002). Since α-MSH has been found to produce anxiogenic effects in some species (Panksepp & Abbott, 1990;Rao et al, 2003) there would be reason to expect α-MSH would increase vocalizing. It may be worth noting in this regard that the subjective impression of the primary observer during Experiment 1 was that α-MSH had a calming effect on the pups.…”

“…Supporting this contention, ICV administration of α-MSH (a melanocortin agonist) enhanced the expression of the gene encoding NUCB2 in the PVN (Oh et al 2006), whereas administration of the melanocortin receptor antagonist, SHU9119, attenuated nesfatin-1-induced satiety (Oh et al 2006). The melanocortin system appears to be involved in the mediation of anxiety-related behaviors, as administration of α-MSH reduced time spent on the open arms of the EPM as well as the number of licks in the Vogel test (Rao et al 2003;Chaki et al 2003). Moreover, pretreatment with SHU9119 dose-dependently attenuated stressor-induced anxiogenic behaviors (measured on the EPM) (Liu et al 2007).…”

“…The number of entries into the open arms, the time spent in open arms as well as the number of closed-arm entries were recorded for 5 min by a trained observer blind to the treatment protocol. It was indicated in numerous analytical studies that the closed-arm entries may be a more reliable index of general activity levels than the total arm entries (Pellow et al 1985;Lister 1987;Dawson et al 1995;Rodgers and Johnson 1995;Rao et al 2003). An entry was registered only when all the four paws of the animal were put on the arm.…”
