Fusion of dipalmitoylphosphatidylcholine vesicles at 4.degree.C

Abstract: Small sonicated dipalmitoylphosphatidylcholine vesicles when incubated at 4 degrees C and high concentrations are shown to fuse completely to vesicles about 700-A diameter in 7 days, and these further fuse to about 950 A diameter vesicles after 3-4 weeks. The 950 A diameter vesicles are spherical, homogeneous, mostly unilamellar, have an internal aqueous space about 10 times that of small vesicles, and are stable for at least 6 months. The 950-A vesicles are characterized by agarose gel chromatography, freeze-… Show more

“…Unilamellar vesicles were prepared from the vortex suspension by ultrasonic irradiation in a cup-horn of a Branson Sonifier W185 (Danbury, Conn.I at above the phase-transition temperature as previously reported (Kamaya et al, 1984). The preparation was aged at 4~ for one week to obtain relatively homogeneous size distribution by fusing vesicles, as described by Wong et al (1982). When observed by electron microscopy after negative staining by 0.5% ammonium molybdate, the aged preparation showed an average diameter of 0,7/xm.…”

“…The preparation was aged at 4~ for two weeks to obtain homogeneous and relatively large vesicle suspension (Wong et al, 1982). Long-chain alkanols were mixed with the vesicle suspension by sonication under the flow of nitrogen gas.…”

“…Fujii et al (1 993) used sonicated vesicles, which may have been composed of phase-separated domains of gel-phase DPPC and fluidphase DOPS. Such vesicles are probably unstable, and will have a greater tendency to undergo fusion and lysis (Nicolussi et al, 1982;Wong et al, 1982). As Fujii et al (1993) pointed out, the cationic and amphipathic nature of both native and reduced defensins may promote fusion and lysis in this system.…”
