Fusarium head blight (FHB) is a serious cereal disease in the Czech Republic. The most important pathogen associated with FBH is Fusarium graminearum, which can produce trichothecenes, mainly deoxynivalenol (DON) and its derivates. A set of 103 F. graminearum isolates were isolated from naturally infected wheat ears collected from 20 localities (25 ears from one locality) within the Czech Republic, in the year 2004. The ears were evaluated for DON content by ELISA. Maximum detected value was 30.7 mg/kg. A group of PCR assays targeting the segments of the Tri7, Tri13 and Tri3 genes were used to determine the chemotypes of F. graminearum isolates. All the isolates belonged to DON producing chemotype. Further discrimination revealed that almost all (99.03%) isolates belonged to 15-ADON chemotype, and only one (0.97%) isolate belonged to 3-ADON chemotype. The genetic variability of the isolates was assessed from their AFLP fingerprints. The populations were highly heterogeneous both within and between locations, and no clear evidence for the association between AFLP profile and geographic origin was found out.

The principal agents of Fusarium head blight in the main cropping area of Argentina were investigated in heavily infected samples. The ability of the isolates to produce trichothecenes was determined by GC and HPLC. Fusarium graminearum was the predominant species and of 33 isolates, 10 produced deoxinivalenol (DON) (0.1-29 mg kg ), 10 produced Fusarenone X (0.1-2.4 mg kg -1 ) and 7 produced zearalenone (0.1-0.6 mg kg -1 ). These results suggest that F. graminearum strains isolated from the wheat growing regions in Argentina belong to DON chemotype. Although some strains produced both deoxinivalenol and nivalenol, nivalenol was produced in lower levels. The natural occurrence of nivalenol in wheat affected by head-blight collected in the main production area during two years (2001)(2002) was also determined. From 19 samples 13 were contaminated with deoxinivalenol in a range of 0.3 to 70 mg kg -1 and 2 samples with both deoxinivalenol (7.5 and 6.7 mg kg -1 ) and nivalenol (0.05 and 0.1 mg kg -1 ), respectively. This is the first report of natural occurrence of nivalenol in wheat cultivate in Argentina.

Trichothecene mycotoxin genotypes ofFusarium graminearum sensu strictoandFusarium meridionalein wheat from southern Brazil

A total of 82 fungal isolates was obtained from wheat kernel samples affected by fusarium head blight collected from 20 locations in southern Brazil. Polymerase chain reaction (PCR) assays were used to characterize trichothecene mycotoxin genotypes [deoxynivalenol (DON), nivalenol (NIV) and two acetylated derivatives of DON]. To identify isolates that producing DON and NIV, portions of the Tri13 gene were amplified. To identify 3-acetyl-deoxynivalenol (3-ADON) and 15-acetyl-deoxynivalenol (15-ADON) genotypes, portions of Tri3 and Tri12 were amplified. Nearly all of the isolates studied (76/82) were of the DON/15-ADON genotype. Six of the isolates were of the NIV genotype. The DON/3-ADON genotype was not observed. Portions of three genes were sequenced from representative isolates of the NIV and DON/ 15-ADON genotypes and compared with sequences from curated reference isolates of Fusarium in GenBank. blast queries for individual gene sequences and pairwise comparisons of percentage identity and percentage divergence based on 1676 bp of concatenated DNA sequence suggested that the isolates representing the DON/15-ADON genotype were Fusarium graminearum sensu stricto and the isolates representing the NIV genotype were Fusarium meridionale . This is the first detailed report of trichothecene mycotoxin genotypes of F. graminearum and F. meridionale in Brazil.
