Functional role of BK virus tumor antigens in transformation

Abstract: We have examined the role of the human papovavirus BK virus (BKV) tumor (T) antigen(s) in the maintenance of transformation and have identified the domain of T antigen essential for transformation. BKV-transformed BHK 21 and NIH 3T3 cells expressing antisense T-antigen RNA lose their ability to grow in soft agar, indicating the need for the continued expression of T antigen for the maintenance of the transformed phenotype. Experiments using translation termination linker insertion and deletion mutagenesis of B… Show more

“…6). The apparent molecular weights were affected little by the deletions, consistent with previous observations made while using the same monoclonal antibody and immunoprecipitation for BKV T-Ag deletion mutations (Simmons et al, 1977;Nakshatri et al, 1988). These reports proposed the explanation that the effects may be due to extensive posttranslational modifications.…”

“…1). The PAbIO8 monoclonal antibody, which recognizes SV4O T-Ag and reacts moderately well with JCV and BKV T-Ags (Nakshatri et al, 1988;BoIlag et al, 1989: Haggerty et al, 1989, was used. As a control, 0.5 pg HpaIdigested pRS V-/I-Gal was used.…”

“…We found this to be true in both our in vivo studies with monkey cell lysates and our in vitro studies with GST fusion proteins of the human retinoblastoma family members, suggesting that the interactions between BKV TAg and both monkey and human pRb family members are the same. Previous studies indicate that BKV TAg is normally expressed at low levels in a variety of host cells, including human embryonic kidney cells (5,62,66,74,76). In addition, BKV DNA levels in normal tissue surrounding urinary tract tumors are lower than in the tumors themselves (59).…”

“…The ATPase domain provides the energy needed for helicase activity. The outside surface of the helicase domain also binds to the tumour suppressor protein p53, thus preventing cell cycle arrest and apoptosis [ 37 , 38 , 39 , 41 ].…”
