Functional Integration of Grafted Neural Stem Cell-Derived Dopaminergic Neurons Monitored by Optogenetics in an In Vitro Parkinson Model

Abstract: Intrastriatal grafts of stem cell-derived dopamine (DA) neurons induce behavioral recovery in animal models of Parkinson's disease (PD), but how they functionally integrate in host neural circuitries is poorly understood. Here, Wnt5a-overexpressing neural stem cells derived from embryonic ventral mesencephalon of tyrosine hydroxylase-GFP transgenic mice were expanded as neurospheres and transplanted into organotypic cultures of wild type mouse striatum. Differentiated GFP-labeled DA neurons in the grafts exhib… Show more

“…Possible targets would be activation and recovery of breathing, suppression of seizure-like activity, amelioration of parkinsonian symptoms, recovery of blindness and optical deep brain stimulation. Yet, peripheral nervous system targets could also be plausible (Nagel et al, 2005;Gradinaru et al, 2009;Adamantidis et al, 2010;Dani et al, 2010;Gradinaru et al, 2010;Zhang et al, 2010;Depuy et al, 2011;Peled, 2011;Tønnesen et al, 2011).…”

“…Albeit these overall positive data, additional long term trials on the safety, efficacy, as well as further understanding of the biological mechanisms activated by graft procedures still need to be accomplished. So far, real functional integration between ectopically grafted SCderived dopaminergic neurons in the denervated striatum has been demonstrated only in organotypic cultures of wild type mouse striatum (Tonnesen et al, 2011). Therefore, studies aimed at the characterization of the molecular basis of the integration in/differentiation of (genetically modified) NSC, and their progeny, within the dopaminergic network deserve further extensive development.…”
