Functional diversity of FGF‐2 isoforms by intracellular sorting

Abstract: Regulation of the subcellular localization of certain proteins is a mechanism for the regulation of their biological activities. FGF-2 can be produced as distinct isoforms by alternative initiation of translation on a single mRNA and the isoforms are differently sorted in cells. High molecular weight FGF-2 isoforms are not secreted from the cell, but are transported to the nucleus where they regulate cell growth or behavior in an intracrine fashion. 18 kDa FGF-2 can be secreted to the extracellular medium wher… Show more

“…(37,38) IRES can also mediate the translation of several protein isoforms with different subcellular localizations: for example, fibroblast growth factor-2 (FGF-2) can be produced as distinct isoforms by the alternative initiation of translation on an IRES-containing mRNA and these isoforms are differently sorted in cells. (39) In most cases described above, ribosomes recognize the alternative TIS by leaky scanning (Fig. 1B).…”

“…We have previously shown that a nuclear isoform of fibroblast growth factor-2 (FGF-2 23 ) directly interacts and co-localizes with SMN (21,22). FGF-2 belongs to the expanding family of growth factors with intranuclear and intracellular localization in addition to their role as extracellular signaling molecules (23).…”
