Fumonisin B1 production by fusarium proliferatum strain M5991 in a modified myro liquid medium

Abstract: Fusarium proliferatum strain M5991 cultures were grown in shake flasks containing modified Myro (MM) medium (MgSO4 reduced to 0.5 g/L) plus 0, 0.25, 0.50, 0.75, 1.00, or 1.25% (v/v) hot-water and corn-hull-extract (CHE) for 69 days. After 4 days of incubation, shake flask liquid cultures with 0.75, 1.00, and 1.25% (v/v) CHE showed a reduction in pH from 6.0 to 2.6 and consumed sucrose at > 6.3 g/L/d. After 69 days of incubation, the same shake flask cultures produced over 7.8 g/L cell mass and over 990 mg/L fu… Show more

“…The column was washed with 20% acetonitrile:80% water containing 0.1% trifluoroacetic acid (TFA), and FB 1 was eluted with 50% acetonitrile:50% water containing 0.1% TFA. The purity of the isolated FB1 was determined by analytical HPLC of the o-phthaldialdehyde (OPA) derivative (Hopmans and Murphy, 1993 Dantzer et al (1996a) by inoculating capped baffled Erlenmeyer flasks containing 500 mL of modified Myro medium with a 4 day shake flask culture of F. proliferatum strain M5991. Fumonisin B 1 was isolated and purified by the procedure as described in Dantzer et al (1996b).…”

“…F. verticillioides MRC 826 corn culture material (after inoculation with 10À20 beads of fumonisin spores, in modified Myro medium (0.5 g/L of MgSO 4 ) plus 1% corn-hull-extract incubated for 69 days) containing 9807 μg of FB1/g was obtained from the Veterinary Medical Diagnostic Laboratory at the University of Missouri, Columbia, MO. 9 Rice culture infected with F. graminearum R6575 (containing 640 μg of deoxynivalenol/g and 57 μg of 15-acetyldeoxynivalenol/g), after inoculation on potato dextrose agar plates and incubation in carboxymethylcellulose media (composed of carboxymethylcellulose, NH 4 NO 3 3 7H 2 O, and yeast extract) mixed with enriched white rice, was prepared previously by Dr. Hendrich's laboratory according to the method of Clifford et al 10 Diet. Rats were divided into five groups: control (fed AIN93G diet obtained from Harlan Teklad, Madison, WI), Grainsure E (0.5% in AIN 93G), toxins (mixture of fumonisin B1, deoxynivalenol, and zearalenone in AIN93G), toxins + Grainsure E, and pair-fed controls (fed daily the mean food intake of toxin group as measured on the previous day).…”

“…Fumonisin production of cultures grown in a modified Myro medium containing 1% (w/v) corn-hullextract [24] for 5 days on rotary shaker (125 rpm) at 25 °C, in the dark was measured by means of the Toxiklon ELISA kit [25].…”
