Full sensitivity of P2times2 purinoceptor to ATP revealed by changing extracellular pH

Abstract: A full pharmacological characterization was carried out on a recombinant ATP-gated ion channel (P2X, purinoceptor) expressed in Xenopus oocytes. This slowly-desensitizing neuronal P2X2 purinoceptor, activated by ATP (EC50=4.6+1 jM at pH 7.4; n=4), showed the agonist potency order: ATP ,2-MeSATP = ATPyS ATPoSS> > Bz-ATP. The receptor affinity for ATP was enhanced 5-10 fold by acidifying the bathing solution (to pH 6.5) but was diminished 4-5 fold in an alkaline solution (pH 8.0).The maximum activity of P2X2 pur… Show more

“…This inhibition can be relieved by increasing extracellular calcium. These results are in contrast to studies on P2X 2 receptors in which responses to ATP may be potentiated by acidification (King et al 1996). The P2X 4 receptor ion channel is thus likely to be functionally modulated by the level of surrounding synaptic activity via local changes in extracellular pH and calcium in a way that differs from other P2X receptors.…”

“…1996) whilst P2X 2 receptor-mediated responses have been shown to be potentiated by acidification (King et al 1996). In the present study we have investigated the effects of altering extracellular pH on a brain-localized P2X receptor (Garcia-Guzman et al 1997), the human P2X 4 receptor, expressed in Xenopusoocytes.…”

“…P2X 2 homomeric receptors show only an increase in peak currents when pH is changed from control (pH 7.4) to a pH of 6.5 [26,45,51]. In addition, pH variations to 5.5 caused an increase in P2X 2 peak currents but a decrease on P2X 2/6 peak currents [27].…”

“…We have shown recently that ATP-evoked excitation of inspiratory neurones underlies carbon dioxide-mediated excitation of respiratory activity . Our observation that P2X 2 reactivity was present in the VLM is therefore particularly signi¢cant, since this receptor subtype is known to be highly sensitive to changes in pH (King et al, 1996).…”
