Full-Range Intracellular pH Sensing by an Aggregation-Induced Emission-Active Two-Channel Ratiometric Fluorogen

Abstract: Intracellular pH (pHi) is an important parameter associated with cellular behaviors and pathological conditions. Sensing pHi and monitoring its changes in live cells are essential but challenging due to the lack of effective probes. We herein report a pH-sensitive fluorogen for pHi sensing and tracking. The dye is a tetraphenylethene-cyanine adduct (TPE-Cy). It is biocompatible and cell-permeable. Upon diffusing into cells, it responds sensitively to pHi in the entire physiological range, visualizing the acidi… Show more

“…However, they suffer from defects of the double bonds: (1) E/Z isomers with different properties in photophysics and sensing are formed during syntheses. The isomers are very hard to separate and thus they are often used in a mixture, leading to a complicated situation of mechanistic understanding; [13] (2) many double-bond containing AIEgens, such as Schiff bases and dicynao-substituted compounds, are vulnerable to nucleophiles, such as CN − , HS − , and OH − , [14] making them chemically unstable; and (3) the double bonds easily undergo photoisomerization and photooxidization upon excitation, leading to low photostability. [12] On the other hand, Type II AIEgens are of well-defined structures with more clear AIE mechanism, better chemical and photostabilities.…”

“…[ 22 ] This unusual phenomenon has been extensively explored and shown a wide range of applications, such as fl uorescence sensors, biological probes, and organic light-emitting diodes (OLEDs). [23][24][25][26][27] It is noteworthy that the morphology and crystallinity of the aggregated state of AIE molecules gave significant effects on their optical and electronic properties. [ 28,29 ] The organic AIE units can be controlled to assemble to various structures such as nanoparticles, nanowires and nanoplates, via different strategies.…”
