Full-Field Optical Coherence Tomography as a Diagnosis Tool: Recent Progress with Multimodal Imaging

Abstract: Full-field optical coherence tomography (FF-OCT) is a variant of OCT that is able to register 2D en face views of scattering samples at a given depth. Thanks to its superior resolution, it can quickly reveal information similar to histology without the need to physically section the sample. Sensitivity and specificity levels of diagnosis performed with FF-OCT are 80% to 95% of the equivalent histological diagnosis performances and could therefore benefit from improvement. Therefore, multimodal systems have bee… Show more

“…6,18 Dynamic FFOCT is also highly sensitive to membrane flickering and cell surface remodeling, as observed in red blood cells and in lymphocytes. 34 We expect the dynamic FFOCT signal to be high in cytoplasm where many organelles are trafficking, in red blood cells, and in photoreceptor IS which are full of mitochondria. The dynamic FFOCT signal in the photoreceptor OS is not so significant but might be caused by membrane fluctuations since membrane density is important and no cytoskeleton is present to prevent thermal fluctuations.…”

“…Recently, a multimodal setup ( Fig. 1(a)) was proposed to combine static and dynamic FFOCT with fluorescence microscopy [40]. This setup can be used to confirm the FFOCT identity of specific cell types or behaviours via comparison with the fluorescent signal.…”

“…Our approach was to implement full-field optical coherence tomography (FF-OCT), or optical coherence microscopy (OCM) [9][10][11][12] using a time domain (TD) set-up -this configuration avoids expensive galvo-mirror scanners [3][4][5]13] and swept sources [14][15][16]. The use of a motorized translation stage and LED sources instead of more expensive super-luminescent diode (SLED) or halogen sources was a key driver in our cost-cutting.…”
