“…Unlike the rapidly fluctuating blood glucose level, the relative amount of HbA1c (HbA1c % ¼ [HbA1c/total hemoglobin] Â 100) reflects the average blood glucose concentration during the past 2 -3 months; and is therefore able to serve as a long-term diagnostic marker [7]. HbA1c is formed by the nonenzymatic reaction of glucose with the N-terminal valine residue of the b-chain of hemoglobin [8,9]. Proteolysis of HbA1c yield fructosyl-valine (Fru-Val), an electro-active compound representative of HbA1c; this reaction sequence is conjectured as involving the following steps: (1) the N-terminal residue of the b-chain in the HbA1c molecule undergoes an enzymatic reaction promoted by an endoproteinase to produce glycated hexapeptides; (2) a protease catalyzes the reaction in which the glycated hexapeptides produce Fru-Val [10], which is used here as a model compound for HbA1c determination.…”
