From Interface to Solution: Integrating Immunoassay with Netlike Rolling Circle Amplification for Ultrasensitive Detection of Tumor Biomarker

Abstract: Both the 3D solution and the 2D interface play important roles in bioanalysis. For the former, reactions can be carried out adequately; while for the latter, interfering substance can be eliminated simply through wash. It is a challenge to integrate the advantages of solution-based assays and the interface-based assays. Here, we report an immuno-NRCA (netlike rolling circle amplification) strategy, which integrates immunoassay with NRCA for the ultrasensitive detection of tumor biomarker, by taking the assay o… Show more

“…This could be attributed to the different absorption and activation of H 2 O 2 on the PtNi‐N‐rGO/GCE at different H 2 O 2 concentrations. [ 39 ] From the calibration curve, the H 2 O 2 sensor detection limit is 2.8 × 10 ‐9 m as determined by 3 Sb −1 ( S is the stand deviation of average blank and b is the slope of the calibration curve [ 21a,40 ] ). The comparisons with other H 2 O 2 detection methods are summarized in Table S1 (Supporting Information).…”

“…Recently, the same group reported an immunoassay based on RCA, named immuno-NRCA, for the ultrasensitive detection of tumor biomarkers. Different from the conventional enzyme linked immunosorbent assay (ELISA), this immuno-NRCA first detected the targets on a 2D interface, and then the reaction was transferred to a 3D solution for signal amplification [ 48 ].…”
