Frequency of HLA-G exon 8 polymorphisms and kidney allograft outcome in Iranian population

Abstract: The 14-bp polymorphism in exon 8 of the HLA-G gene is associated with HLA-G mRNA stability and the patterns of alternative isoform splicing and may influence the functionality of the HLA-G molecule. HLA-G expression was related to allograft acceptance and fewer episodes of acute rejection during heart, kidney and liver-kidney transplantation. In order to determine a possible correlation between the 14-bp insertion/deletion polymorphism and kidney allograft outcome in our population, genomic DNA was isolated fr… Show more

“…No statistically significant differences were found between the HLA-G 14 bp deletion/insertion allele and genotypes between kidney transplant rejection group and non-rejection group. This observation is in agreement with data reported by others [29,30]. In the group with non kidney graft rejection higher frequencies of homozygous -14/-14 bp genotype were observed in comparison to recipients with acute rejection, as confirmed by others [21,22].…”

“…Moreover, HLA-G 14 bp insertion/deletion polymorphism (rs16375) was amplified by polymerase chain reaction (PCR) [21] with the forward primer HLAG1 5′-GTGATGGGCTGTTTAAAGTGTCACC-3′ and the reverse primer HLAG2 5′-GGAAGGAATGCAGTTCAGCATGA-3′.…”

“…Included characteristics were the name of the first author, year of publication, country of study origin, source of control group, sample size, and HardyWeinberg equilibrium (Table 1). [4][5][6][7][8][9] Statistical analyses All statistical analyses were performed with software (Stata 11.0, StataCorp, College Station, TX, USA) using 2-sided tests. The chi-square test was used to compare genotype (likelihood ratio P value) and allele frequencies (continuity correction P value) between the kidney recipient and healthy control groups.…”

“…Therefore, 5 studies that included a total of 907 kidney transplant recipients (rejection, 271 patients; no rejection, 636 patients) and 887 healthy control subjects were included in the meta-analysis (Table 1). [5][6][7][8][9] According to the Newcastle-Ottawa quality assessment scale, all the studies were of high quality ( Table 2).…”
