In this study, antibacterial activity of crude extract from Orthosiphon stamineus was analyzed. Extraction through immersing method was performed using methanol and aqueous extraction from whole plant (except root). Spot test and MIC was conducted for both extracts to screen for their antibacterial activity against Staphylococcus aureus and 10 methicillin resistant Staphylococcus aureus (MRSA). No antibacterial activity was observed for all strain tested in the spot test assay. However, observation through MIC test showed that both extracts have antibacterial activity. The aqueous extract showed antibacterial activity in the range of 6.25 mg/mL to 50 mg/mL against S. aureus, MRSA strains (ATCC 4330, TM, WM2, K31, K32, K39, and K52). The antibacterial activity of methanol extract was found to be in the range of 12.5 mg/mL to 50 mg/mL). Methanol extract was found to be more active against S. aureus and 9 MRSA strains (ATCC 4330, BM3, TM, WM2, K31, K32, K39, K52 and K59. The aqueous extract tested on S. aureus has the highest MIC value which is 6.25 mg/mL. In conclusion, O. stamineus was found to have potential as an anti-MRSA agent. Further analysis has to be done in order to develop a novel anti-MRSA drug to combat multiple resistant bacteria.

Objective: In the gastrointestinal system, gastric ulcer is one of the common serious problems in human life and gives contribution against morbidity and mortality incidence. The pathophysiology of gastric ulcer is an imbalance between aggressive factor and mucosal integrity factor. Increase of aggressive factors and decrease of mucosal integrity factors have potential against developed of gastric ulcer disease. The objective of the research was to evaluate antioxidant and gastric ulcer healing effect of Orthosiphon stamineus (Benth.) leaves extract in aspirin-induced rats. Methods:In vivo antiulcer activity of Orthosiphon leaves extract was evaluated through several parameters involves gastric acidity, number of ulcers, diameters of ulcers, ulcer index (UI), and healing ratio. Dose level of Orthosiphon leaves extract which used in this study such as 250 and 500 mg/kg, respectively. Antioxidant activity of Orthosiphon leaves extract was evaluated by 1,1-diphenyl-2-picrylhydrazyl hydrate (DPPH) method. Histopathological of the stomach was performed using hematoxylin-eosin stained. Results:The results of the study showed that groups which given Orthosiphon leaves extract have significantly different for gastric ulcer healing compared to the control group and were supported by histopathological analysis. The Orthosiphon leaves extract also showed maximum scavenging activity at a concentration of 100 µg/ml (58.86% inhibition) and minimum at 50 μg/ml (29.60% inhibition) with inhibitory concentration 50% (IC 50 ) 84.54 µg/ml when compared to ascorbic acid as the standard with IC 50 5.08 µg/ml by DPPH method. Conclusion:It can be concluded that from the experimental study of O. stamineus (Benth.) leaves extract has potential antiulcer activity in aspirininduced rats model and antioxidant effect using DPPH method. Stomach tissues regeneration in gastric ulcer model might be affected by the improvement of antioxidant status.

This study evaluates the antibacterial effects of Zingiber officinale essential oil and Orthosiphon stamineus water extract against Enterococcus faecalis. The herbs were prepared in various concentrations to determine their minimum inhibitory concentrations (MIC) and growth inhibitory effect. Anti-adhesion activities of the herbs were determined by co-incubation with E. faecalis cultures for 6 and 24 h. Biofilm disruption activities were determined by adding the studied herbs into preformed E. faecalis biofilm. The effects on the morphology of E. faecalis grown as biofilm were studied using scanning electron microscopy (SEM). The MICs of ginger oil and O. stamineus extract were 0.31 and 25 mg/mL, respectively. Between the tested herbs, ginger exhibited greater inhibitory effects on the growth of E. faecalis grown in suspension mode. Both herbs generally showed anti-adhesion activities in inverse concentration-dependent manner. No significant biofilm disruption activities by both herbs were observed. SEM analyses showed E. faecalis cell surface changes in the treated biofilm. The studied herbs may have compromised the integrity of the bacterial cell membrane. These findings suggest that the studied herbs may have better antibacterial activities against E. faecalis in suspension mode compared to biofilm mode, with ginger oil showed greater antibacterial activity compared to O. stamineus extract.
