Fractal Analysis of Skeletal Muscle Tissue of Rats Subjected to Stretch Injury

Abstract: SUMMARY:The aim of this study was to analyze changes in the skeletal muscle tissue of rats after muscle stretch injury using fractal analysis. Nineteen rats were divided into three groups, Control (n= 5) (C), Lesion two days (n= 7) (L2), and Lesion seven days (n= 7) (L7). Histological slides of the gastrocnemius muscle of the animals were prepared and stained with Hematoxylin and eosin for analysis of the inflammatory process, and picrosirius for the demonstration of collagen. The stained slides were subsequen… Show more

“…In this way FD has been successfully used to identify cancer cells, discriminate the architecture of muscles with dystrophy as well as differentiate brain cells (Lee et al, 2014;Pantic et al, 2015Pantic et al, , 2014Cury et al, 2018), and these processes are linked to the number of nuclei in the analyzed site. In another study from our group (Ozaki et al, 2015), FD proved to be effective in differentiating injured muscle from healthy muscle, a fact that was again observed in the present study. Although, the box-counting method founded differences in FD of a healthy muscle to an injured in histological aspects, but it was not sensitive to differentiate two distinct treatments or to find differences in collagen levels.…”

“…In the fractal dimension when we submitted HE-stained blades to the binarization process, the nuclei are detached from the rest of the cells, so it is possible to measure the size of the infiltrate or other findings by the analysis of its fractal dimension [25] The findings of this study show that DF is a tool that can be used as a means of quantitative analysis of a lesion, which adds to the study an independent evaluation of the evaluator. Although there were not many significant statistical differences in the comparison between the groups, the fractal dimension analysis showed that animals poisoned by venom and even those submitted to treatment with antiofidic serum showed an increase in the fractal dimension in comparison to CG animals, suggesting that they have a larger fractal dimension.…”

“…However it is known that this type of analysis is dependent on the analyzer, which leads to intra and interobserver variations [21]. Unlike this method, fractal analysis, in addition to quantifying histological changes, is independent of the evaluator, since it is an automatic analysis performed by software [25].…”

“…Cuts measuring 5 μm in thickness were made in a cryostat microtome at −20 °C, placed on slides and stained with hematoxylin-eosin (HE) for analysis of the structure of the muscles, and Picrosirius red for collagen analysis. 20 , 21 …”
