Formation of Malonaldehyde from Phospholipid Arachidonate during Microsomal Lipid Peroxidation

Abstract: The thiobarbituric acid reacting material produced during enzymatic microsomal lipid peroxidation has been identified as malonaldehyde. The malonaldehyde was condensed with urea to form 2‐hydroxy‐pyrimidine, which was identified by its ultraviolet spectrum, chromato‐graphic properties, and mass spectrum. Incubations with phosphatidyl choline labelled with tritiated arachidonate yielded 2‐hydroxy‐pyrimidine with a specific activity nearly equal to that of the phospholipid arachidonate. Incubations with tritiate… Show more

“…The activities of non-enzymatic antioxidant such as reduced glutathione (GSH) and enzymatic antioxidants such as reduced glutathione peroxidase (GPx), SOD, CAT and thiobarbituric acid reactive substances (TBARS) were assayed according to the methods of Ellman (1959), Rotruck et al(1973), Kakkar et al(1984), Sinha (1972) and Niehans and Samuelsson (1968), respectively. The protein content in the tissue homogenate was estimated by following the protocol of Lowry et al(1951).…”

“…C [21], Vit. E [22] and lipid per oxidation in terms of TBARS [23], as the case may be in serum, liver, heart and kidney and tri acyl glycerol (TAG) in adipose tissue by standard methods. Histopathology of liver tissues was carried out as per the procedures followed before [9].…”
