Formas de esterilização do GA3 e reação morfogênica em microestacas de mamoeiro

Abstract

RESUMO -Este trabalho objetivou estudar os efeitos do ácido giberélico (GA 3 ) sobre o alongamento de brotos de mamoeiro 'Tainung 01' em meio de multiplicação in vitro e posterior enraizamento ex vitro. Brotações com menos de 1 cm de comprimento foram submetidas ao alongamento em meio MS contendo GA 3 , nos seguintes tratamentos, em mg L -1 : T 1 = 0,0; T 2 = 0,5 esterilizado junto com o meio de cultura; T 3 = 0,5 esterilizado a frio; T 4 = 2,0 esterilizado junto com o meio de cultura; T 5 = 2,0 esterilizado a frio. Após esses tratamentos, as brotações foram submetidas ao enraizamento ex vitro. Observou-se que o GA 3 nos níveis 0,5 e 2,0 mg L -1 , utilizado após autoclavagem em meio de multiplicação, não se mostrou efetivo para o alongamento dos ramos, mas quando esterilizado por filtragem, mostrou-se efetivo no alongamento das brotações; no entanto, é prejudicial no estádio subsequente de enraizamento, podendo, estes, necessitar de novo subcultivo em meio suplementado com auxina, visando a induzir o enraizamento. FORMS OF STERILIZATION OF GA 3 AND MORPHOGENIC REACTION IN MICROCUTTINGS OF PAPAyAABSTRACT -This study investigated the use of gibberellin (GA 3 ) in medium in vitro propagation of papaya 'Tainung 01' and checking the elongation of shoots and their behavior in the ex vitro rooting.. Shoots less than one cm were subjected to elongation on MS medium containing GA 3 treatments as in mg L -1 : T1 = 0.0 T2 = 0.5 with sterilized culture medium, T3 = 0.5 sterilized cold, T4 = 2.0 with sterilized culture medium, T5 = 2.0 sterile cold. After these treatments, the shoots were subjected to ex vitro rooting. It was observed that GA 3 at levels of 0.5 and 2.0 mg L -1 used in autoclaving after the multiplication has not proved effective for lengthening the shoots, but when sterilized by filtration proved to be effective in lengthening the shoots, however, is harmful in the subsequent stage of rooting and can latter, require subculture to fresh medium supplemented with auxin to induce rooting.
