“…The mechanism appears to be partially related to suppression of Th2 responses and enhancement of Th1 responses, as production of IL-4, IL-5, and IL-13 was significantly reduced with treatment, while IFN-γ production increased (103). In the murine model, suppression of anaphylactic reactions lasted at least six months after treatment (104,105).…”

“…Indeed, our data show that 80% of mice reached a clinical score of ϳ3.5 and a very robust ϳ8°C drop in core body temperature. We did perform a number of detailed experiments using previously reported protocols and explored a series of methodological variations including: an increase in the number of oral sensitizations to up to eight, executing two oral challenges rather than one (17,21), and increasing gut permeability and absorption of Ag with ethanol (22) at the time of challenge. However, we were not able to elicit consistent anaphylactic reactions not only in C57BL/6 mice but also in C3H/HeJ mice.…”

“…We then extended the study and found that FAHF-2-treated mice were completely protected from anaphylaxis following challenge for up to 5 wks post-therapy, and that reduced IgE levels remained significantly lower at 5 wks posttherapy. Complete protection was confirmed using several well-established parameters, including anaphylactic symptom scores, decreased core body temperatures, elevated plasma histamine levels [32] and vascular leakage. [33] This therapeutic effect was associated with immunoregulatory effects on Th1-Th2 responses.…”

“…We named the refined formula FAHF-2. A preliminary study [32] found that, as with FAHF-1, FAHF-2 also completely blocked peanut-induced anaphylaxis. We then extended the study and found that FAHF-2-treated mice were completely protected from anaphylaxis following challenge for up to 5 wks post-therapy, and that reduced IgE levels remained significantly lower at 5 wks posttherapy.…”
