The objective of this study was to compare the effectiveness of the protocols for superstimulation of follicular growth in Thai native heifers. Heifers (n = 20) were randomly divided into four groups of five heifers/group. Heifers were given a single dose by i.m. administration of 100 mg Follicle Stimulating Hormone dissolved in polyvinylpyrrolidone (FSHp) at 24 h. Ovum pick up (OPU) occurred at 72 h (F24O72 protocol; Group 1) or 96 h (F24O96 protocol; Group 2), and at 36 h and OPU at 72 h (F36O72 protocol; Group 3) or 96 h (F36O96 protocol; Group 4) after follicular ablation. The dynamics of ovarian follicular growth were monitored by twice-daily ultrasonographic examinations. Blood sample collections were performed every 12 h after initiation of treatment for assessment of FSH, E2 and P4 profiles. All heifers were subjected to eight repeated sequential sessions of OPU. The follicular deviation commenced 24±5.32 h after follicular ablation in all groups. The circulatory FSH surged quickly from 24 to 36 h (>0.8 ng/ml) after follicular ablation and circulatory estrogen levels steadily increased from 36 h until OPU in all groups. At the end of the OPU sessions, the mean number of aspirated follicles/heifer/session in F36O72 protocol (Group 3) and F36O96 protocol (Group 4) were higher than in the two other groups (p<0.05). The number of cumulus-oocyte complexes (COCs), cleaved and day 8 blastocysts rates in the F36O72 protocol (Group 3) were higher than in the other groups (p<0.05). It can be concluded that a single dose i.m. administration of 100 mg FSHp at 36 h and OPU at 72 h after follicular ablation (F36O72 protocol; Group 3) was the most effective protocol for superstimulation of follicular growth for repeated OPU and subsequent in vitro embryo production in Thai native heifers.

The aim of this study was to assess the dose (300 to 600 IU) effects of equine chorionic gonadotropin (eCG) on the preovulatory follicle diameter, growth rate and time of ovulation characterized by echography. The eCG was injected at the end (D 0 ) of the 7-day treatment with a controlled internal device release (CIDR®) and a PGF 2α being injected 2 days before the removal of the CIDR® (d −2 ). The 120 N'Dama female were distributed into five experimental groups. The control group (n=26) was treated with physiological saline at the removal of the CIDR®, while the animals in the four treated groups received, respectively, 300 IU (n=25), 400 IU (n=24), 500 IU (n=22) and 600 IU (n=23) of eCG. The diameter of the preovulatory follicle was significantly higher (P<0.05) in the animals treated with 300 IU (10.1±1.4 mm) than in untreated animals (9.3±1.2 mm). Follicle growth rate was significantly (P<0.05) higher in treated animals (1.0± 0.4 mm/day) than in the control group (0.9±0.4 mm/day). The average interval between the time of eCG injection and ovulation was similar in the non-treated (83.7±14.4 h) and treated animals (79.7±11.9). Treated animals showed a significant increase in the percentage of ovulation (94.7 % compared to 73.1 %) (P<0.01). Use of eCG contributed towards synchronising the time of ovulation between 72 to 96 h, which would facilitate the use of systematic insemination.

Oocytes recovered were selected and those viable were submitted to in vitro embryo procedures. Data were analyzed by ANOVA, using PROC MIXED, in which time of OPU, replicate and interaction between these factors were considered as fixed effects, and donor was considered as a random effect. Higher blastocyst rate (6 days after IVF), number of blastocysts (9 days after IVF), and number of nuclei of hatched embryos were produced when OPU was done on D2. However, OPU on D6 also produced a higher blastocyst rate (6 days after IVF), and did not differ from other times of OPU concerning number of nuclei of hatched embryos. In Chapter 2, two experiments were performed. In Experiment 1, the effect of 50 mg of injectable progesterone was evaluated in the synchronization of follicular wave emergence of Nelore, Holstein, and buffalo heifers submitted to a progestin plus 2 mg of estradiol benzoate protocol (n=10/ group). Data were analyzed by logistic regression, by PROC GLIMMIX. The time of follicular wave emergence was not affected by administration of injectable progesterone. Besides, follicular wave emergence occurred between 4.2 and 4.5 days after the beginning of synchronization protocol for all genetic groups. In Experiment 2, the effects of time of OPU relative to follicular wave emergence (1, 3 or 5 days) and of genetic group (Nelore, Holstein, and buffalo) on in vitro embryo production were evaluated. A total of 27 heifers (9 of each genetic group, assigned in the 3 times of OPU) were submitted to 6 OPU sessions, in a cross-over design. Oocytes recovered were selected and those viable were submitted to in vitro embryo procedures. Data were analyzed by ANOVA, using PROC MIXED, in which time of OPU, genetic group, replicate and interaction between these factors were considered as fixed effects, and donor was considered as a random effect. Results demonstrated that Nelore heifers showed a better efficiency for in vitro embryo production than Holstein or buffalo heifers. Additionally, OPU performed at different times of synchronized follicular wave did not influence in vitro embryo production.
