Fluvoxamine?A new serotonin re-uptake inhibitor: First clinical and psychometric experiences in depressed patients

Abstract: The clinical efficacy of fluvoxamine (DU 23000)--the first selective serotonin re-uptake inhibitor of the new class of 2-aminoethyloximethers of aralkylketones was investigated in endormorphous depressed patients during 5 weeks treatment with mean daily dosages of approximately 150 mg DU 23000. A marked and statistically significant improvement of the overall and detailed psychopathology was noted by means of the Global Clinical Impression and Hamilton Rating Scale as early as in the first week of drug adminis… Show more

“…The these studies confirm the safety and tolerance as well as the predicted antidepressant effects (Doogan, 1980;Saletu et al, 1977;Wright & Derlber, 1978;Klok et al, 1981;Freeman et al, 1981). The maximum doses of fluvoxamine and clovoxamine prescribed in human experiments have been 400 mg/day.…”

“…While only one clinical case of fluvoxamine-induced torsade de pointes has been reported (Manet et al, 1993), bradycardic action (de Wilde et al, 1983;Granfors et al, 2004;Pacher and Kecskemeti, 2004;Saletu et al, 1977), QT-interval prolongation (Klok et al, 1981;Nia et al, …”

“…Although in vitro chronotropic or inotropic effect of fluvoxamine has not been reported, in vivo negative chronotropic effect of fluvoxamine has been observed in dogs (Roos, 1983) and patients (de Wilde et al, 1983;Granfors et al, 2004;Pacher and Kecskemeti, 2004;Saletu et al, 1977), and in vivo negative inotropic effect has been shown in rabbits (Claassen, 1983;Wouters and Deiman, 1983). Meanwhile, fluvoxamine decreased the total peripheral resistance, leading to the hypotensive action.…”
