Fluorometric Riboflavin Titration in Plasma by Ribof lavin-Binding Apoprotein as a Method for Vitamin B2 Status Assessment

Abstract: A method for plasma riboflavin determination by riboflavin-binding apoprotein titration has been proposed for vitamin B2 status evaluation. The method is based on the formation of riboflavin-apoprotein complex accompanied by full loss of fluorescence peculiar to free riboflavin. The data obtained have demonstrated a correlation with indicators of the vitamin B2 status such as urinary excretion, erythrocyte content, and stimulation of the glutathione reductase activity by flavin adenine di… Show more

“…Official methods for quantification of Rf in food samples are based usually on HPLC measurement and microbiological assays. Alternative methods involve electrochemical or fluorescent characteristics of flavins for standard solutions, [1][2][3] biological fluids, 4,5 and pharmaceutical tablets 6 but none for food matrixes. In addition to these approaches, methods based on biological properties of Rf through the binding with Rf binding protein (RBP) from egg white [7][8][9] have been investigated.…”

“…Vitamin status was assessed by the content of vitamins in the serum. The concentration of retinol (vitamin A) and betacarotene was determined using high performance liquid chromatography [12], riboflavin (vitamin B 2 )by fluorimetric method using riboflavin-binding apoprotein [13], 25-hydroxyvitamin D [25(OH)D] (vitamin D)enzyme immunoassay method using the test system «ELECSYS Vitamin D Total» (F. Hoffmann-La Roche Ltd., Switzerland). Persons with indicators that did not reach the lower limit of the norm [13] were considered to be insufficiently provided with vitamins.…”
