New rapid, sensitive and simple electrometric method was developed to determine sertraline hydrochloride (Ser-Cl) in its pure raw material and pharmaceutical formulations. Membrane sensors based on heteropolyacids as ion associating material were prepared. Silicomolybdic acid (SMA), silicotungstic acid (STA) and phosphomolybdic acid (PMA) were used. The slope and limit of detection are 50.00, 60.00 and 53.24 mV/decade and 2.51, 5.62 and 4.85 μmol L−1 for Ser-ST, Ser-PM and Ser-SM membrane sensors, respectively. Linear range is 0.01–10.00 for the three sensors. These new sensors were used for the potentiometric titration of Ser-Cl using sodium tetraphenylborate as titrant. The surface morphologies of the prepared membranes with and without the modifier (ion-associate) were studied using scanning and atomic force microscopes.

Pancuronium bromide, 1,1Ј-(3a,17b-diacetoxy-5a-androstan-2b,16b-ylene)bis (1-methyl-piperidinium) dibromide [15500-66-0] is general anaesthesia used in surgery for muscle relaxation and as an aid to endotracheal intubation 1) (Chart 1). Several methods of determination of this drug have been reported in the literature, including high performance liquid chromatography (HPLC), 2-6) capillary gas chromatography using nitrogen sensitive detection, 7) liquid chromatography-mass spectrometry 8-12) spectrophotometric, [13][14][15] fluorimetric, 16,17) and mass spectrometric, 18) methods. A potentiometric ion selective electrode method based on pancuronium-tetraphenylborate and pancuronium-dipicrylaminate has been reported. 19,20) The aim of this study is to describe the construction, performance characteristics of pancuronium modified carbon paste electrodes based on the pancuronium-tetraphenylborate (Panc-TPB) and pancuronium-reineckate as electroactive material and dibutyl phthalate as solvent mediator and the analytical applications of these sensors in determination of pancuronium bromide in pharmaceutical preparation and biological fluids in batch and flow injection conditions. ExperimentalReagents and Materials All chemicals were of analytical grade. Double distilled water was used throughout all experiments. Pure grade pancuronium bromide and the pharmaceutical preparation Bromurex Ampoule (2 mg/ml) were provided by Sedico Pharmaceutical Co., 6 October City, Egypt. Sodium tetraphenylborate (NaTPB), ammonium reineckate, dioctyl sebacate (DOS), tricresyl phosphate (TCP), tetradodecylammonium tetrakis-(4-chlorophenyl)borate (ETH 500) were obtained from Fluka (U.S.A.), dibutyl phthalate (DBP) and dioctyl phthalate (DOP) from Merck (Germany). Graphite powder (1-2 micron) from Aldrich (U.S.A.).Apparatus Potentiometric and pH measurements were carried out using a Seibold G-103 digital pH/mV meter (Vienna, Austria). A Techne circulator thermostat Model C-100 was used to control the temperature of the test solutions. A new, simple, fast, reproducible and sensitive reversed phase HPLC method, using a new stationary phase containing embedded urea polar groups, has been developed and validated for the simultaneous determination of clobutinol hydrochloride (CLO) and doxylamine succinate (DOX) in syrups. The determination was carried out on a C8 urea column (125 mm x 3.9 mm i.d., 5 mm particle size) synthetized at the Liquid Chomatography Laboratory (LabCrom) of the Chemistry Institute of Unicamp. The mobile phase consisted of a mixture of acetonitrile:methanol:phosphate buffer (pH 2.5) in the gradient mode. The diode array detector (DAD) was operated at 230 nm for CLO and 262 nm for DOX. The method showed adequate precision, with relative standard deviations (RSD) less than 1%. The presence of the excipients did not interfere in the results of the analysis. Accuracy was determined by adding standards of the drugs to a placebo and good recovery values were obtained. The analytical curves were linear (r 2 0.9999 for CLO and 0.9998 for DOX) over a wide concentration range (2.4-336 μg mL -1 for CLO and 2.3-63 μg mL -1 for DOX). The solutions were stable for at least 72 hours at room temperature. The criteria for validation using the ICH guidelines were fulfilled.Uniterms: Clobutinol hydrochloride/determination. A determinação foi realizada em uma coluna C8 uréia (125 mm x 3,9 mm d.i., 5 mm tamanho de partícula) sintetizada em nosso laboratório (LabCrom). A fase móvel consistiu de mistura de acetonitrila:metanol:tampão fosfato pH 2,5, em eluição por gradiente. O detector por arranjos de diodo (DAD) foi utilizado a 230 nm para CLO e a 262 nm para DOX. O método apresentou precisão adequada, com desvio padrão relativo menor que 1%. A presença de excipientes não interferiu nos resultados obtidos. A exatidão foi realizada pela adição dos padrões dos fármacos ao placebo e valores de recuperação aceitáveis foram obtidos. As curvas analíticas mostraram-se lineares (r 2 0,9999 para CLO e 0,9998 para DOX) em uma ampla faixa de concentração (2,4-336 μg mL -1 para CLO e 2,3-63 μg mL -1 para DOX). A solução padrão foi estável por 72 horas a temperatura ambiente. Os parâmetros de validação foram realizados conforme o guia ICH.
