Flow cytometric study on cell kinetics of brain tumours and their cultured cells

Abstract: With the aid of flow cytometry (FCM), distribution of DNA content in 40 cases of brain tumour, primary culture cell, and secondary culture cell can be determined and chronological change after subculture is studied from the analysis of their cell cycle. In most primary cultures, proliferating index (PI) is likely to decrease, which suggests that environmental change might affect the growth activity. In comparison with that of the original sample, DNA-histogram of the secondary culture can be divided into the f… Show more

“…Moreover, LIs of cultured anaplastic astrocytomas and glioblastomas have been found to be lower than the values obtained in cultures of low-grade astrocytomas. Our in vitro cell kinetic results are, however, in agreement with reports of other workers (20)(21)(22).…”

“…More recently, the estimation of various cell kinetic parameters have provided yet another index for more accurate malignancy assessment (6-12). However, very few studies are available on the morphological appearance ( 13-1 3 , GFAP expression (2, 13, [16][17][18][19] and growth kinetics (20)(21)(22)(23) of cultured human astrocytoma cells and no report, to the best of our knowledge, is available correlating all these parameters in vitro with the grade of malignancy of the parent tissue.…”
