Bombesin (BBN), an analog of gastrin-releasing peptide (GRP), specifically binds to GRP receptors, which are overexpressed in human prostate cancer (PC). Here, we synthesized a BBN-modified gadolinium oxide (Gd 2 O 3 ) nanoprobe containing fluorescein (Gd 2 O 3 -5(6)-carboxyfluorescein [FI]-polyethylene glycol [PEG]-BBN) for targeted magnetic resonance (MR)/optical dual-modality imaging of PC. The Gd 2 O 3 -FI-PEG-BBN nanoparticles exhibited a relatively uniform particle size with an average diameter of 52.3 nm and spherical morphology as depicted by transmission electron microscopy. The longitudinal relaxivity (r 1 ) of Gd 2 O 3 -FI-PEG-BBN (r 1 =4.23 mM −1 s −1 ) is comparable to that of clinically used Magnevist (Gd-DTPA). Fluorescence microscopy and in vitro cellular MRI demonstrated GRP receptor-specific and enhanced cellular uptake of the Gd 2 O 3 -FI-PEG-BBN in PC-3 tumor cells. Moreover, Gd 2 O 3 -FI-PEG-BBN showed more remarkable contrast enhancement than the corresponding nontargeted Gd 2 O 3 -FI-PEG according to in vivo MRI and fluorescent imaging. Tumor immunohistochemical analysis further demonstrated improved accumulation of the targeted nanoprobe in tumors. BBN-conjugated Gd 2 O 3 may be a promising nanoplatform for simultaneous GRP receptor-targeted molecular cancer diagnosis and antitumor drug delivery in future clinical applications.

Background: Glucose transporter (Glut), a cellular transmembrane receptor, has a key role in the metabolism of cell glucose and is also associated with various human carcinomas. Objective: In this study, we evaluated a magnetic resonance (MR) imaging contrast agent for tumor detection based on paramagnetic gadolinium oxide (Gd2O3) coated polycyclodextrin (PCD) and modified with glucose (Gd2O3@PCD-Glu) for the targeting of overexpressed glucose receptors. Material and Methods: In this experimental study, 3T magnetic resonance imaging (MRI) scanner was used to assess the specific interactions between Glut1-overexpressing tumor cells (MDA-MB-231) and Gd2O3@PCD-Glu NPs. Furthermore, the capacity of transporting Gd2O3@PCD-Glu NPs to tumor cells was evaluated. Results: It was found that the acquired MRI T1 signal intensity of MDA-MB-231 cells that were treated with the Gd2O3@PCD-Glu NPs increased significantly. Based on the results obtained, Gd2O3@PCD-Glu NPs can be applied in targeting Glut1-overexpressing tumor cells in vivo, as well as an MRI-targeted tumor agent to enhance tumor diagnosis. Conclusion: Results have shown that glucose-shell of magnetic nanoparticles has a key role in diagnosing cancer cells of high metabolic activity.

Graphene oxide (GO) is a bidimensional novel material that exhibits high biocompatibility and angiogenic properties, mostly related to the intracellular formation of reactive oxygen species (ROS). In this work, we set up an experimental methodology for the fabrication of GO@peptide hybrids by the immobilization, via irreversible physical adsorption, of the Ac-(GHHPH)4-NH2 peptide sequence, known to mimic the anti-angiogenic domain of the histidine-proline-rich glycoprotein (HPRG). The anti-proliferative capability of the graphene-peptide hybrids were tested in vitro by viability assays on prostate cancer cells (PC-3 line), human neuroblastoma (SH-SY5Y), and human retinal endothelial cells (primary HREC). The anti-angiogenic response of the two cellular models of angiogenesis, namely endothelial and prostate cancer cells, was scrutinized by prostaglandin E2 (PGE2) release and wound scratch assays, to correlate the activation of inflammatory response upon the cell treatments with the GO@peptide nanocomposites to the cell migration processes. Results showed that the GO@peptide nanoassemblies not only effectively induced toxicity in the prostate cancer cells, but also strongly blocked the cell migration and inhibited the prostaglandin-mediated inflammatory process both in PC-3 and in HRECs. Moreover, the cytotoxic mechanism and the internalization efficiency of the theranostic nanoplatforms, investigated by mitochondrial ROS production analyses and confocal microscopy imaging, unraveled a dose-dependent manifold mechanism of action performed by the hybrid nanoassemblies against the PC-3 cells, with the detection of the GO-characteristic cell wrapping and mitochondrial perturbation. The obtained results pointed out to the very promising potential of the synthetized graphene-based hybrids for cancer therapy.
