Trionyx sinensis hemorrhagic syndrome virus (TSHSV) is a pathogen that causes severe hemorrhagic syndrome and irreversible damage to different infected tissues of Pelodis cus sinensis, ending in the death of affected organisms. In the present study, the histopathological characteristics of TSHSV-infected P. sinensis were analyzed and compared by HE staining. Relative and absolute quantification (iTRAQ)-based proteomic analysis was employed to explore the molecular pathology of liver injury. Anatomical features indicated that TSHSV caused obvious congestion in the liver, kidney, intestine, and other tissues of P. sinensis. The typical clinical symptoms included hepatomegaly, fragility, spotty and severe congestion in liver tissue, and also obvious intestinal bleeding. The histopathological studies corroborated such lesions in the liver and kidney, etc. iTRAQ-based proteomic analysis revealed that there were 252 differentially expressed proteins in the liver tissue between healthy and infected P. sinensis, of which 118 proteins were upregulated and 134 proteins were downregulated. GO enrichment analysis and KEGG pathway analysis initially revealed the molecular mechanism of pathological changes in P. sinensis by TSHSV infection. The expression of some differentially expressed proteins was further confirmed by qRT-PCR. These results provided important information for the pathological diagnosis of TSHSV-caused disease, as well as the mechanism underlying TSHSV-caused disease.

RESUMO: A entrada de Helicoverpa spp. no Brasil resultou em profundas mudanças bioecológicas de espécies, favorecendo a evolução da resistência da praga à inseticidas e variedades Bt. Será necessário o desenvolvimento de um plano efetivo de manejo integrado da praga, que engloba estratégias de manejo da resistência e táticas de controle, como inseticidas e variedades Bt. O objetivo deste trabalho foi avaliar a eficiência de Bacillus thuringiensis (Bt) no controle da lagarta Helicoverpa spp. na cultura da soja (Glycine max), comparada com os principais inseticidas químicos do mercado. Foi realizado um experimento de campo, na Fazenda Leila, na safra 2015/2016, com a cultivar de soja TMG 132 RR, com infestação natural dos insetos-praga. O experimento foi montado em um delineamento em blocos ao acaso, com parcelas de 5 linhas, com 5 metros de comprimento e espaçamento de 0,45 metros entre linhas, onde foi deixada uma distância de 0,9 metros entre cada parcela totalizando 20 parcelas. Os inseticidas testados foram: Clorantraniliprole (Prêmio®) 0,15 L ha-1 , Indoxacarbe (Avatar® 0,50 L ha-1) e Clorfenapir (Pirate®) 1,00 L ha-1 e o bioinseticida Dipel® 1,00 L ha-1. As aplicações iniciaram após levantamento, no dia anterior, considerando a quantidade média de Helicoverpa spp., quando verificou-se a presença de 4,75 lagartas por tratamento considerando todos os instares. Para as avaliações, foram realizadas contagens de lagartas no 1º dia, 2º dia, 4º dia e 9º dia após a aplicação. Considerou-se a quantidade de Helicoverpa spp. vivas por parcela, sendo contabilizadas lagartas neonatas ao 2º instar, 3º e 4º instar e acima de 5º instar, realizando-se quatro avaliações após a aplicação. O tratamento com Dipel, na dosagem de 1,00 L ha-1 apresentou a mesma eficácia dos demais inseticidas testados para o controle de Helicoverpa spp, na cultura da soja, até o 9º dia após aplicação do produto. Chinese soft-shelled turtle (Pelodiscus sinensis) is commercially cultured in East and Southeast Asia for its nutritional and medicinal values. In this study, we identified interleukin-1β (IL-1β) from Chinese soft-shelled turtle. The full-length cDNA of Pelodiscus sinensis IL-1β (tIL-1β) consists of 1529 base pairs with an 831-base-pair open reading frame, encoding 277 amino acids. The guanine-cytosine (GC) content in the coding sequence and 3’ untranslated region of tIL-1β is considerably higher than that of other vertebrates. Its mRNA expression level increased significantly during Aeromonas hydrophila infection. The tIL-1β lacks the typical IL-1β-converting enzyme (ICE) cut site found in mammalian IL-1β, but still could be cleaved by turtle caspase-1. By mutating the potential cleavage sites, we identified aspartic acid (Asp/D) 130 as the ICE cut site in tIL-1β. The peptide truncated at D130 was expressed using the baculovirus expression system; its bioactivity is confirmed by the ability to induce cyclooxygenase-2 (COX-2) and tIL-1β itself in peripheral blood monocytes. In conclusion, we characterized IL-1β from Chinese soft-shelled turtle and identified its D130 as a non-typical ICE cut size.
