Fungal diseases caused by native pathogens and pathogens introduced with planting stock have a significant impact on exotic plantation forestry in the tropics. Teratosphaeria destructans (formerly Kirramyces destructans) is a serious pathogen causing leaf, bud and shoot blight diseases of Eucalyptus spp. in plantations in the sub-tropics and tropics of south-east Asia. This pathogen was first discovered in Indonesia in 1995 and has subsequently spread to Thailand, China, Vietnam and East Timor. The biology, ecology and genetics of this important pathogen have not been explored yet. The objective of this study was, thus, to determine the genetic diversity and movement of T. destructans throughout south-east Asia using multi-gene phylogenies and microsatellite markers. Out of nine gene regions only two microsatellite markers detected a very low nucleotide polymorphism between isolates; seven other gene regions, ITS, β-tubulin, EF1-α, CHS, ATP6 and two microsatellite loci, reflected genetic uniformity. The two polymorphic molecular markers resolved six haplotypes among isolates from Indonesia and only a single haplotype elsewhere in Asia. The low diversity observed among isolates in the region of the first outbreak is as expected for a small founder population. The spread of a single clone over large distances throughout the region supports the hypothesis of spread via the human-mediated movement of germplasm.

The Eucalyptus stem canker pathogen Teratosphaeria zuluensis was discovered in South Africa in 1988 and it has subsequently been found in several other African countries as well as globally. In this study, the population structure, genetic diversity and evolutionary history of T. zuluensis were analysed using microsatellite markers to gain an enhanced understanding of its movement in Africa. Isolates were collected from several sites in Malawi, Mozambique, Uganda and Zambia. Data obtained were compared with those previously published for a South African population. The data obtained from 334 isolates, amplified across eight microsatellite loci, were used for assignment, differentiation and genetic diversity tests. STRUCTURE analyses, θ st and genetic distances revealed the existence of two clusters, one dominated by isolates from South Africa and the other by isolates from the Zambezi basin including Malawi, Mozambique and Zambia. High levels of admixture were found within and among populations, dominated by the Mulanje population in Malawi. Moderate to low genetic diversity of the populations supports the previously held view that the pathogen was introduced into Africa. The clonal nature of the Ugandan population suggests a very recent introduction, most likely from southern Africa.

ABSTRACT:The gall wasp, Leptocybe invasa, is a devastating pest of eucalypt plantations, woodlots, and urban trees in the Middle East, the Mediterranean basin, Africa, India, South-East Asia, and China. It was first observed in Israel in 2000 and has since spread rapidly south and east. It reached southern Vietnam in 2002 and has since moved northward devastating nurseries and young plantations. Eighteen species of eucalypts were selected for a sentinel trial in northern Vietnam to determine their susceptibility to pathogens in the region. They were also rated for the impact of Leptocybe invasa. Leptocybe invasa was capable of feeding and forming galls on 13 species from six sections within Eucalyptus and one species of Corymbia. The impact was greater in the nursery. Five of the Eucalyptus spp. which were susceptible in the nursery were not damaged in the field. The most susceptible hosts in Vietnam were E. camaldulenis, E. grandis, and E. tereticornis, both in the nursery and field trial. Variation in the susceptibility of provenances of E. camaldulensis, E. urophylla, and E. grandis were observed, which is promising for breeding for resistance and long-term control.
