Environmental Factors Associated with the Presence of Vibrionaceae in Tropical Cage‐Cultured Marine Fishes

This study investigated the environmental factors associated with the presence of Vibrionaceae in economically important cage‐cultured tropical marine fishes: the Asian Seabass Lates calcarifer, snapper Lutjanus sp., and hybrid grouper Epinephelus sp. Fish sampling was conducted at monthly intervals between December 2016 and August 2017. The body weight and length of individual fish were measured, and the skin, eye, liver, and kidney were sampled for bacterial isolation and identification. Water physicochemical parameters during the sampling activities were determined, and the enumeration of total Vibrionaceae count was also conducted from water and sediment samples. Nine species of Vibrio were identified, including V. alginolyticus, V. diabolicus, V. harveyi, V. campbellii, V. parahaemolyticus, V. rotiferianus, V. furnissii, V. fluvialis, and V. vulnificus. Photobacterium damselae subsp. damselae was also identified. A total of 73% of the isolated Vibrio belonged to the Harveyi clade, followed by the Vulnificus clade (5.5%) and Cholera clade (0.6%). Highest occurrence of Vibrio spp. and P. damselae subsp. damselae was found in hybrid grouper (72%), followed by Asian Seabass (48%) and snapper (36%). The associations of Vibrio spp. and P. damselae subsp. damselae with the host fish were not species specific. However, fish mortality and fish size showed strong associations with the presence of some Vibrio spp. On average, 60% of the infected cultured fish exhibited at least one clinical sign. Nevertheless, inconsistent associations were observed between the pathogens and water quality. The yearlong occurrence and abundance of Vibrionaceae in the environmental components indicate that they might serve as reservoirs of these pathogens.

In the aquaculture industry, the application of probiotics is well known widely used to control disease, improve water quality and reduce demands for the use of antibiotics or disinfectants. However, some local gut bacteria can bind to harmful bacterial component. For better efficacy of probiotics in the digestive tract of carnivorous fish, encapsulation can be a simple, harmless, and improved method to maintain the microflora of the digestive tract thereby enhancing the immune system of fish. This study was conducted to investigate the effects of encapsulated probiotic Bacillus sp. D2.2.on gut bacterial communities and immune system in brown-marbled grouper Epinephelus fuscoguttatus. One hundred fish weighing about 40 g were divided randomly into five groups including negative control and positive control groups which were fed by a commercial diet only and a diet containing probiotic respectively, the third to fifth groups were fed by 1, 2, and 3 g/kg encapsulated probiotics dose of feed respectively. The results showed that encapsulated probiotic increase viability of probiotic bacteria and also affect the abundance of lactic acid bacteria. Immune response of the brown-marbled grouper also increased significantly after the application of encapsulated probiotics.

Antibacterial Effect of Unripe Grape, Lemon and Pomegranate Juice Against Vibrio Parahaemolyticus in Mussels Samples

Bu çalışmada, koruk, limon ve nar suyunun Vibrio parahaemolyticus'a karşı antibakteriyel etkisi araştırılmıştır. Bu amaçla, midye örneklerine yaklaşık 10 7 KOB/g V. parahaemolyticus inokule edilmiştir. İnokülasyon yapılan midye örnekleri 0, 30, 60 ve 90 dk koruk, limon ve nar suyunda bekletilmiş ve 4 o C'da 6 gün boyunca depolanmıştır. Depolama periyodunda, V. parahaemolyticus sayısı, pH ve titre edilebilir asitlik belirlenmiş ve duyusal parametreler açısından değerlendirilmiştir. Araştırma sonucunda, en yüksek antibakteriyel etki koruk suyu uygulanan deneme gruplarında görülmüştür (P <0.05). Denemenin 0. gününde koruk, limon ve nar suyu uygulanan gruplardaki V. parahaemolyticus sayısı, konsantrasyona bağlı olarak sırasıyla 2.22-2.64; 1.58-1.85 ve 1.44-1.71 log KOB/g düzeyinde azalma göstermiştir. Bu azalma eğilimi depolamanın diğer günlerinde de devam etmiştir. Duyusal değerlendirme sonucunda, nar suyunun, midye örneklerinin renk ve görünüşünü olumsuz etkilediği belirlenmiştir (P <0.05). Araştırma sonunda, ev uygulamalarında kullanılan sofra tipi asitleştiricilerin, özellikle koruk suyunun yüksek düzeyde antibakteriyel etkisinin olduğu, su ürünlerinde gıda güvenliğin sağlanması için kullanılabileceği belirlenmiştir.
