Final Safety Assessment of Thiodipropionic Acid and Its Dialkyl Esters as Used in Cosmetics

Abstract: Dilauryl thiodipropionate (DLTDP), dicetyl thiodipropionate, dimyristyl thiodipropionate, distearyl thiodipropionate, and ditridecyl thiodipropionate are dialkyl esters of their respective alcohols and thiodipropionic acid (TDPA) used in cosmetics. Ingested DLTDP was excreted in the urine as TDPA. Single-dose acute oral and parenteral studies and subchronic and chronic repeated dose oral studies did not suggest significant toxicity. Neither DLTDP nor TDPA was irritating to animal skin or eyes and they were not… Show more

“…These findings further buttress the literature suggesting that the magnitude of the extractables and leachables problem is growing in both size and scope and affecting many diverse industries. 14,15 In conclusion, we report the identification of three leachables from disposable plastic labware that were found to be MAO-B-specific inhibitors, among which was the antioxidant DLTDP. Bioactive contaminants in drug discovery assays pose many challenges, including costly loss of time, productivity, and efficiency due to investigating the source of interfering agents and resolving conflicting data sets.…”

“…The earlier study of Kunwar et al [53] have documented the biological applications of hentriacontane such as antimicrobial, cancer treatment, acne control etc., from Bauhinia variegate and Vitex negundo. The bioactive component (2), distearyl thiodipropionate which is a diester of stearyl alcohol and thiodipropionic acid was used as an antioxidant [54]. The bioactive component (3), 1, 2-benzenedicarboxylic acid, mono (2-ethylhexyl) ester was reported earlier as a secondary metabolite, possessing antimicrobial, antioxidant and anti-proliferative activities [55][56][57].…”

“…However, when the water content is high, as in the preparation of creams under acidic conditions, the ester bonds in both GMS and isopropyl myristate become broken, which leads to their break down into various degradation products. Accordingly, isopropyl myristate breaks down into isopropanol isopropyl myristate and myristic acid, while GMS decomposes into stearic acid and glycerol [11,12]. Based on this decomposition, the stability of anti-aging cream preparations can be evaluated.…”
