Fibroblast Growth Factor 21 Reverses Hepatic Steatosis, Increases Energy Expenditure, and Improves Insulin Sensitivity in Diet-Induced Obese Mice

Abstract: OBJECTIVE-Fibroblast growth factor 21 (FGF21) has emerged as an important metabolic regulator of glucose and lipid metabolism. The aims of the current study are to evaluate the role of FGF21 in energy metabolism and to provide mechanistic insights into its glucose and lipid-lowering effects in a high-fat diet-induced obesity (DIO) model. RESEARCH DESIGN AND METHODS-DIOor normal lean mice were treated with vehicle or recombinant murine FGF21. Metabolic parameters including body weight, glucose, and lipid levels… Show more

“…Circulating FGF21 and adiponectin levels were markedly increased in the blood from Mfn2‐adKO mice compared to control mice, linked to a 30‐fold higher FGF21 expression in BAT from Mfn2‐adKO mice. Circulating FGF21 is positively correlated with liver insulin sensitivity, and FGF21 administration prevents hepatic steatosis under HFD condition (Xu et al , 2009). This great upregulation of FGF21 in BAT might be consequent to mitochondrial dysfunction, as recently proposed (Kim et al , 2013).…”

“…With this regard, irisin may manipulate PPAR-ɑ, which is an instrumental regulator in fat metabolism that modifies fat oxidation through a thermogenesis mechanism (42). Additionally, PPAR-ɑ regulates fibroblast 21 growth factor (FGF21), which can ameliorate liver osteatosis and insulin sensitivity (43). Therefore, irisin can regulate the amount of intra-liver triglyceride through FGF21.…”

“…Notably, FGF21 has been shown to reverse hepatic steatosis in animal models of NAFLD and insulin resistance (Camporez et al, 2013b;Xu et al, 2009). In particular, Camporez et al showed that these improvements were associated with reduced hepatocellular triglyceride and diacylglycerol contents, decreased activation of PKCε in the liver, as well as improved insulin signaling (Camporez et al, 2013b).…”
