FGF-2 Stimulates Periodontal Regeneration

Abstract: The efficacy of the local application of recombinant human fibroblast growth factor-2 (FGF-2) in periodontal regeneration has been investigated. In this study, a randomized, double-blind, placebo-controlled clinical trial was conducted in 253 adult patients with periodontitis. Modified Widman periodontal surgery was performed, during which 200 µL of the investigational formulation containing 0% (vehicle alone), 0.2%, 0.3%, or 0.4% FGF-2 was administered to 2- or 3-walled vertical bone defects. Each dose of FGF… Show more

“…Fibroblast growth factors (FGFs) have shown potential effects on the repair and regeneration of a wide spectrum of tissues, including skin, blood vessel, muscle (Tassi et al 2011), de novo adipogenesis (Tabata et al 2000), tendon/ligament (Hankemeier et al 2005), cartilage (Martin et al 2001), bone (Kimura et al 2008), tooth (Kitamura et al 2011), and nerve tissues (Yun et al 2010).…”

“…Additionally, the topical application of bFGF was shown to significantly induce PDL regeneration without ankylosis, root resorption and epithelial down-growth in an experimental alveolar bone defect model in dogs (Murakami et al, 2003). Moreover, the regenerative effect of bFGF on PDL tissue in patients with periodontitis was confirmed in a randomized controlled phase II clinical trial and a multi-center randomized clinical trial (Kitamura et al, 2011).…”

“…Growth factors commonly utilized in regenerative dentistry include bone morphogenetic proteins (BMPs), platelet-derived growth factor (PDGF), enamel matrix derivative (EMD), and fibroblast growth factor-2 (FGF2) [17][18][19][20][21]. Despite attempts that have been made to combine grafting materials or barrier membranes with recombinant growth factors, autogenous bone has remained the gold standard of bone grafting materials due to its excellent combination of osteogenesis, osteoinduction, and osteoconduction.…”
