Ferroptosis: a new strategy for Chinese herbal medicine treatment of diabetic nephropathy

Abstract: Diabetic nephropathy (DN) is a serious microvascular complication of diabetes. It has become a leading cause of death in patients with diabetes and end-stage renal disease. Ferroptosis is a newly discovered pattern of programmed cell death. Its main manifestation is the excessive accumulation of intracellular iron ion-dependent lipid peroxides. Recent studies have shown that ferroptosis is an important driving factor in the onset and development of DN. Ferroptosis is closely associated with renal intrinsic cel… Show more

“… 89 Mounting studies suggest that ferroptosis plays an important role in kidney cells injury and is one of the key mechanisms to induce DN, supporting that ferroptosis could be a potential therapeutic target for DN. 90 , 91 The marker of ferroptosis, GPX4, is decreased in db/db mice and T2D patients, which means ferroptosis plays a pathophysiologic role in DN, 68 which means ferroptosis might serve as a target for treating DN. In this section, we mainly separately discuss the role of ferroptosis in podocytes, GECs, GMCs and TECs.…”

“…Over the past few decades, the way people with diabetic nephropathy are treated has changed dramatically (Sawaf et al, 2022), with many patients with refractory disease benefitting from new treatments but a large proportion of patients still progress to advanced disease stages. The pathogenesis of diabetic nephropathy is complex, involving oxidative stress, an inflammatory response, autophagy, programmed cell death and other mechanisms (Duan et al, 2021;Wan et al, 2022;Han et al, 2023;Wei et al, 2023).…”
