Feasibility of Fecal MicroRNAs as Novel Biomarkers for Pancreatic Cancer

Abstract: IntroductionPancreatic cancer (PCA) is an aggressive tumor that associates with high mortality rates. Majority of PCA patients are diagnosed usually at late tumor stages when the therapeutic options are limited. MicroRNAs (miRNA) are involved in tumor development and are commonly dysregulated in PCA. As a proof-of-principle study, we aimed to evaluate the potential of fecal miRNAs as biomarkers for pancreatic cancer.Materials and MethodsTotal RNA was extracted from feces using Qiagen's miRNA Mini Kit. For miRN… Show more

“…miRNome, or miRNA fingerprints), made of both upand down-regulated miRNAs. The PDAC miRNome has been extensively profiled and distinctive miRNA signatures have been shown to be useful in detecting this malignancy in fresh [11] and formalin-fixed paraffin embedded (FFPE) tissues [12], as well as in blood [13], bile [14], biopsy [15], and stool [16] samples.…”

“…As a result, miRNAs may function as regulatory molecules and act as tumor suppressors or oncogenes (9). Among these microRNAs, miR-21 was highly expressed in various types of cancer, including breast, lung and pancreatic cancer (10,11). Functional studies have revealed that miR-21 is an oncogene, which is important in enhancing cell proliferation, promoting cell cycle progression and increasing anti-apoptotic activation in cancer cells (12).…”

“…Available research literature suggests that higher or lower expression of miR-216a-5p occurs in a variety of cancers. Some studies have shown that the expression levels of miR-216a-5p is downregulated in colorectal cancer (16) and pancreatic cancer (17), while others have reported upregulated levels of miR-216a-5p in prostate cancer (18) and liver cancer (19). Intracellular environments of different cells were diversiform, so the signaling pathways of miR-216-5p were described in certain types of tumor.…”

“…Previous studies have demonstrated that miR-216a-5p is dysregulated in a number of malignancies, such as colorectal cancer, pancreatic cancer, prostate cancer and liver cancer (16)(17)(18)(19). However, the expression of miR-216a-5p requires quantification and the role of miR-216a-5p in RCC has not been investigated to date.…”
