“…1) but maximal activity was only recorded after 45-60 min. This confirms previously reported data that are in the same range (30 min) for the closely related desert locust, S. gregaria (Ogoyi et al 1998) and in the fruit beetle after injection of its endogenous AKH peptide Melme-CC (30-45 min, Auerswald et al 2005). Activation of lipase in decapitated moths (M. sexta), however, is much faster: maximum activity is already reached 10 min after injection of the endogenous peptide Manse-AKH (Arrese et al 1996).…”

“…This beetle uses carbohydrates and proline as major flight substrates and lipase activation is necessary for the resynthesis of proline (Ga¨de and Auerswald 2002;Auerswald et al 2005). For locusts, only fragments of information on TAG lipase have been published, such as ideal in vitro assay conditions for lipase from the fat body of L. migratoria (Hirayama and Chino 1987) or, differences in lipase activation in gregarious versus solitary phases in the desert locust, Schistocerca gregaria (Ogoyi et al 1998). Most of the knowledge on activation of TAG lipase in the fat body of locusts derives from the indirect measurement of its action, via monitoring the concentrations of lipids in the haemolymph.…”

“…In addition, the midgut lipase is responsible for generation of DAG from the intestinal TAG pool. Lipase has been investigated in different insect tissues including midgut and fat body (Gilbert et al, 1965;Tietz and Weintraub, 1978;Male and Storey, 1981;Arrese and Wells, 1994;Ogoyi et al, 1998), and its role in fat body during adipokinetic hormone-induced lipolysis is well understood (Ryan and Van der Horst, 2000;Arrese et al, 2001a). Conversely, PAPase has been analyzed, to our knowledge, only in fat body (Hirano and Gilbert, 1967).…”

“…Activity of TAG lipase has been observed in several insect orders, but the data are not comparable due to the different, and sometimes unsuited, methods used (see Downer, 1985). Even data for fat body of locusts (Ogoyi et al, 1998;Hirayama and Chino, 1987) and moths Wells, 1994, 1997;Arrese et al, 1996Arrese et al, , 1999) that were obtained using methods based on that of Hirayama and Chino (1987), vary to a large degree and do not allow inter-specific comparisons. In the present study, we measured, therefore, resting activities of TAG lipase from the fat body of males from three insect species whose flight muscles oxidize different fuels for flight (see Introduction).…”

“…Endogenous AKH peptides have been shown to mobilize lipids in those insects that make use of the lipophorin system (van der Horst et al, 2001;Arrese et al, 2001); it was further demonstrated that the respective AKHs activate a TAG lipase (Arrese and Wells, 1994;Ogoyi et al, 1998). Endogenous AKHs stimulate proline synthesis in the fat body of those insects that use the alanine/acetyl-CoA system (Ga¨de, 1999;Auerswald and Ga¨de, 1999b) but it is not known which enzyme(s) are being activated by the AKHs.…”
