Family-Based Association Study of Lithium-Related and Other Candidate Genes in Bipolar Disorder

Abstract: These results suggest novel candidates and 1 gene (DISC1) previously associated with schizophrenia that merit further study in bipolar disorder. However, polymorphisms in major lithium-signaling genes do not appear to contribute substantially to bipolar liability.

“…A link between sialyltransferase 4A and bipolar disorder has been reported previously. 27 We also identified seven SNPs with Pp3.33 Â 10 À5 located near DRD1 on chromosome 5q35 (Figure 3). DRD1 has a role in behavior and psychiatric disorders, including schizophrenia, 28 autism, 29 attentiondeficit-hyperactivity disorder, 30 nicotine dependence 31 and alcohol dependence.…”

“…These authors suggested that polymorphisms in DISC1 contribute to variations in the psychotic features of bipolar spectrum disorder. DISC1 may represent a new target for the development treatments and diagnostic tools for bipolar disorder 41 . DISC1 is associated with lowered biological activity of ERK (extracellular regulated kinase), reduced grey matter volume in the brain and increased risk for major depressive disorder 43 .…”

“…An analysis of these articles associated identified five genes as the most cited in the literature: CANAC1C [16][17][18][19][20][21][22][23][24] , DAOA [25][26][27][28][29][30][31][32] 22 . Some articles related the genetic polymorphism specifying the classification of TBI 8,25,27,32,35,36,39,41 , in addition to TBI and II 37 . The population studied by the authors of the articles are mostly from ocidente 18,20,22,26,28,30,32,[34][35][36][37][38] and a lower part are the oriente 25,27,33,43 .…”
