Factors affecting mesenchymal stromal cells yield from bone marrow aspiration

Abstract: Objective: This study was to investigate the variables in bone marrow harvesting procedure and individual donor factors which can potentially affect the yield of mesenchymal stromal cells (MSC).Methods: We determined the yield of MSC from bone marrow under different clinical conditions by comparing the MSC colony numbers from: (1) donors of different ages; (2) healthy donors and patients with leukemia; (3) bone marrow aspirated at different time points during marrow harvesting; (4) bone marrow harvested by dif… Show more

“…MSC can be isolated from diverse adult tissues, including bone marrow stroma [4]. The number of MSC in the bone marrow might be reduced by diseases such as osteoporosis [5], leukemia [6], or lung and breast cancers [7]. Aging has also been proposed as an underlying cause of MSC deficiency in the bone marrow; however, this is still a matter of debate.…”

“…Aging has also been proposed as an underlying cause of MSC deficiency in the bone marrow; however, this is still a matter of debate. While a decline in MSC number in bone marrow stroma has been linked to increased donor age in several papers [8, 9], others report the absence of any correlation between the two factors [6, 10]. The reasons for exhaustion of MSC in the bone marrow are still debated; however, oxidative stress has been shown to be one of the major factors influencing tissue aging [11, 12], and as such merits in-depth investigation.…”

“…However, autologous bone marrow procurement is not without its limitations; it is a painful and invasive procedure that is associated with significant morbidity [33]. Moreover, deriving MSCs from bone marrow yields low cell numbers upon processing (approximately 1 MSCs per 10 5 aspirated bone marrow cells) [34, 35]. Generously assuming, a concentration of 3×10 7 nucleated cells per mL of bone marrow aspirate [36] would result in approximately only 600 MSCs per mL of marrow.…”

“…As mentioned previously, approximately only 0.001 % of all bone marrow stromal cells aspirated have been shown to hold multipotent properties [34]. From a practical standpoint, this yields approximately 50–675 MSCs per mL of bone marrow aspirate [35]. In contrast, anywhere from 1×10 5–6 ASCs have been routinely obtained from 1 mL of lipoaspirate [39].…”

“…Alternative harvesting sites are femoral heads (from total hip arthroplasty of an elderly patient population), sternum and less frequently vertebrae. The commonly applied preparation method for the generation of BM-MSC is density gradient centrifugation, but other methods rely on their properties to adhere to plastic [1,[61][62][63][64][65][66] . Ad-MSC are isolated from lipoaspirates obtained during liposuction, lipoplasty or lipectomy procedures.…”
