Fabrication of submicron proteinaceous structures by direct laser writing

Abstract: In this paper, we provide a characterization of truly free-standing proteinaceous structures with submicron feature sizes depending on the fabrication conditions by model-based analysis. Protein cross-linking of bovine serum albumin is performed by direct laser writing and two-photon excitation of flavin adenine dinucleotide. We analyze the obtainable fabrication resolution and required threshold energy for polymerization. The applied polymerization model allows prediction of fabrication conditions and resulti… Show more

“…Several studies involving low photoinitiator concentrations have demonstrated that the threshold protein concentration required for fabrication increases dramatically 32,33,68 . Fabrication without photoinitiators has also been reported 32,33,49,68 , although some attempts were unsuccessful 48 . Such discrepancies in prior works indicate that controlling all the major factors such as laser properties, irradiation conditions, solvent, photoactivator and protein concentration that can affect the outcome of the process is the key to achieving repeatability.…”

Phragma dialysis can increase protein concentrations, but can only be applied for several repetitions due to the physical take-off volume limit.…”

“…Protein molecules in aqueous solution can be cross-linked to construct 3D proteinaceous microstructures by the exposure of a focused femtosecond laser along its trajectory in well-resolved and controlled manners [7][8][9]. Such microstructures made of protein have been applied for cell culture scaffolds [10,11], pH-actuators [9,12,13] and optical elements [13,14].…”
