Fabrication of Hydrogel with Cell Adhesive Micropatterns for Mimicking the Oriented Tumor-Associated Extracellular Matrix

Abstract: For mimicking the fibrous extracellular matrix (ECM), a facile method for patterning anticell adhesive substrate was novelly applied on agarose hydrogel. Without using masks or templates for etching, we applied the magnetic field-induced colloidal assembly of magnetic nanoparticles on the flat agarose hydrogel to form cell-adhesive micropatterns. Meanwhile, tuning the hydrogel substrate's modulus to fit real tissue was experimentally demonstrated. Magnetic nanobeads were also assembled on this hydrogel surface… Show more

“…Both results show that the PLA sample with assembled film of nanoparticles has much higher viability for the bone marrow cells than the plain PLA sample (Figs 2e and f). This may result from the adhesion sites-like effect of the nanoparticles, which was also found in our other work [23,24]. To confirm the reinforcement of cellular adhesion in the presence of nanoparticles, the expression of integrin and cadherin which are important proteins related to cellular adhesion was measured by q-PCR (Fig.…”

“…[32][33][34] Li et al fabricated a series of topographical screens to investigate the physical neural-guidance cues. Hippocampal neurons cultured on anisotropic topographies (linear, circular, angular gratings, triangular, and rectangular) expressed better axonal elongation and dendritic extensions in contrast to those cultured on the isotropic topographies (dots, grids, and squares).…”

“…To introduce the magnetic coupling between building blocks, the magnetic field‐directed assembly was employed to fabricate the linear assemblies of nanoparticles and enhance the magnetic interaction between the nanoparticles. This hybridized hydrogel was reported to be capable of mimicking biological tissues more better in morphology and mechanic property owing to the presence of linearly aligned hard materials . Here, this novel magnetic hydrogel experimentally exhibited the enhanced thermogenesis and the controllable magnetothermal behavior dependent upon direction of the external field with respect to the assembled chains.…”
