Fabrication of a nano-biocatalyst for regioselective acylation of arbutin

Abstract: A nano-biocatalyst has been successfully prepared via coordination between zinc ion and the nitrogen atoms of the amide groups in the Candida sp. 99-125 lipase. The as-prepared nanoparticle has an excellent hierarchical structure with a large number of separate petals. In regioselective acylation of arbutin, the nano-biocatalyst showed enhanced enzyme activities to about 4.3-folds than the free enzyme. Meanwhile, the prepared nano-biocatalyst exhibited good operational reusability and only about 6% of enzyme a… Show more

“…Phosphate (PBS, pH = 7.13) buffer for ADH HNFs was prepared by dissolving 0.56 g Na 2 HPO 4 ·12H 2 O, 0.14 g KH 2 PO 4 , 0.098 g NaCl, and 0.017 g KCl in ultrapure water, respectively, and fixing the volume to 100 mL [ 49 ]. To prepare ADH/Zn 3 (PO 4 ) 2 hybrid nanoflowers, 6 mg of ADH was dissolved in 5 mL of PBS buffer.…”

“…Thus, most DPEases in the active form might be enriched or locked during immobilization, which may be the main reason for the enhanced enzyme activity of the nanoflower. Furthermore, the prepared nanoflower has high surface-to-volume ratios, which can decrease the mass transfer limitations dramatically [33,34], and subsequently improve the enzyme performance.…”

“…Scanning electron microscope (SEM) (S-4800, HITACHI) was employed to examine the morphology of hNFs. 1 H NMR spectra and 13 CNMR were recorded through a Brucker DPX 300 spectrometer at 500 MHz and 126 MHz, respectively. NMR experiment was performed in deuterated chloroform (CDCl 3 ) containing a proper concentration of the sample, a total of 128 scans was recorded.…”

“…Compared with chemical method, enzymatic synthesis of fine chemicals and medicine has been attracting great attention in recent years because of its mild reaction condition, higher catalytic efficiency and the inherent selectivity (10)(11)(12). Of course, in biocatalysis, the immobilized enzymes have various advantages in continuous practice (13) and are attracting more attentions than the free one. For example, the immobilized Candida antarctica lipase B (CALB) has been used to catalyze the synthesis of vitamin A palmitate (14).…”
