Extraction of IgE-Binding Components of Helicobacter pylori by Immunoblotting Analysis in Chronic Urticaria Patients

Abstract: Background: An association between Helicobacter pylori and chronic urticaria has been suspected previously. An IgE-mediated pathway might be a possible link between H. pylori infection and chronic urticaria, and therefore we wanted to prepare an optimal H. pylori antigen to detect H. pylori-specific IgE antibodies in chronic urticaria patients. Methods:H. pylori antigen extracts were prepared in different ways to find the optimal antigen extract to be used in the assays. Immunoblotting was used to detect IgE-b… Show more

“…The direct effects of HP, including immunoglobulins, HP virulence genes, autoimmune reactions, have been considered in CSU development. Immunoglobulin (Ig) G, IgA, and IgE antibodies against HP antigens were detected in some studies . The release of proinflammatory cytokines, granulocyte attraction, as well as B‐ and T‐cell–mediated responses, is elevated in HP‐positive patients, which may all contribute to the development of urticarial lesions .…”

“…Although several studies have been performed to identify immunogloblin-binding antigenic structures of H. pylori, 13,14 little is known about the antigenic structures that are responsible for CIU. Another possible mechanism is the increase of gastric 15,16 Recognition of food allergens may occur more easily in gastric mucosa infected with H. pylori.…”

“…Mice immunised with inactivated herpes simplex develop IgE-specific antibodies to the virus [109]. High levels of IgE are also observed in man following recurrent herpes simplex infection [110] and human IgE antibodies are also known to interact with herpes family viruses including HSV-1 and 2 and the Epstein-Barr and cytomegalovirus [111] and also to C. pneumoniae, H. pylori, and B. burgdorferi [ [112][113][114][115]. IgE-related allergic responses are also involved in C. neoformans infection [116].…”
