Extraction of Betanin Using Aqueous Two-Phase Systems

Abstract: The extraction efficiency of betanin as a natural pigment with antioxidant features is significant. The main reason for studying the different aqueous two-phase systems (ATPSs) for betanin extraction is that the selection of an optimum process of separation is possible. In order to develop an efficient method for the extraction of betanin, ATPS composed of polymer and sodium sulfate was investigated. Sodium sulfate is a strong salting-out agent, which has an important effect on the processes of the extraction … Show more

“…The concentrations of the components at the working point (9% PEG + 8% K 2 HPO 4 + 5% KH 2 PO 4 + 78% H 2 O) in the two-phase region were specified. According to our prior studies, and based on the standards such as (i) ease of two-phase separation, (ii) easy sampling, and (iii) the working points were selected so that both volumes of top and bottom phases are equal . The concentrations of the components existing in the biphasic system were determined as mass fraction.…”

“…Equation was used to estimate the partition coefficient of nisin, which is shown as the nisin mass fraction in the top phase (PEG-rich phase) per mass fraction of nisin in the bottom phase (salt-rich phase) …”

“…According to our previous experimentation and respecting the standards (like the possibility of the easy separation of two phases, simple sampling, and the equal volumes of the top and bottom phases), the working points within the liquid-liquid area were chosen. All the concentrations of the constituents utilized in the system have been calculated on the basis of mass percentage 28,38 .…”

“…In order to purify the target biomolecule in the ATPSs containing nanoparticles and without nanoparticles, the details of the vanillin partitioning process has been given in the Supporting Information. The absorbance of vanillin in the top and bottom phases were determined through spectrophotometry (UV/vis Model: sp-2100uv, USA) at the wavelength of 280 nm where the maximum absorption of vanillin occurred 20 .…”
