Extracellular Matrix in Aging Aorta

Abstract: The aging population is booming all over the world and arterial aging causes various age-associated pathologies such as cardiovascular diseases (CVDs). The aorta is the largest elastic artery, and transforms pulsatile flow generated by the left ventricle into steady flow to maintain circulation in distal tissues and organs. Age-associated structural and functional changes in the aortic wall such as dilation, tortuousness, stiffening and losing elasticity hamper stable peripheral circulation, lead to tissue and… Show more

“…Generally, the structure and the amount of elastin in different types of blood vessels depends on their location in the circulatory system and local hemodynamic conditions. The contribution of elastic structures to vascular biomechanics has been exhaustively studied for decades [ 18 , 20 , 40 , 44 , 53 , 68 ], but new papers are constantly being published [ 15 , 65 , 69 , 70 , 71 , 72 , 73 , 74 , 75 ] showing that there is still room for exploration in this area.…”

“…The extracellular matrix (ECM) determines the structure–function relationships of the aorta, and therefore maintains its homeostasis. ECM is pivotal for a healthy performance [ 24 ]. Age-associated remodeling of the ECM structural components, such as fragmentation of elastic fibers and excessive deposition and crosslinking of collagens, ends in functional stiffening of the aorta ( Figure 2 ) [ 24 ].…”

“…The walls of the arteries become thicker and stiffer. They also lose their elasticity and therefore are more prone to age-related comorbidities, such as hypertension ( 97 ). These structural changes also reflect changes in the composition of extracellular matrix, e.g., less elastin more collagen ( 97 ).…”

These structural changes also reflect changes in the composition of extracellular matrix, e.g., less elastin more collagen ( 97 ). Little is known about the potential role of extracellular matrix's components on initiating or suppressing an autoimmune response.…”
