Objective. In this study, we screened out a type of differentially expressed circular RNA in infantile hemangioma (IH) cells and analyzed the mechanism in the malignant biological behavior of IH. Methods. Based on the GSE98795, GSE100682, and GSE43742 datasets, differential expression analysis of circRNAs, microRNAs, and mRNAs was performed. The relative expression level of RNA was detected by quantitative real-time polymerase chain reaction (qRT-PCR). MTT assay, Transwell, flow cytometry analysis, and western blot were used to study the effects of hsa_circ_0003570, hsa-miR-138-5p, and RGS5 on the proliferation and apoptosis of hemangioma endothelial cells (HEMECs). Results. The hsa_circ_0003570 and RGS5 mRNA were upregulated in HEMECs, but hsa-miR-138-5p was downregulated. Silencing of hsa_circ_0003570 inhibited the proliferation of HEMECs and promoted the apoptosis of HEMECs. The malignant biological behaviors of hsa_circ_0003570 on the proliferation and apoptosis of HEMECs were reversed by hsa-miR-138-5p. Hsa_circ_0003570 acted as the ceRNA of hsa-miR-138-5p and upregulated the expression of RGS5. Silencing of RGS5 inhibited the proliferation, migration, and invasion of HEMECs and promoted apoptosis. Conclusion. Hsa_circ_0003570 promotes IH cell proliferation and inhibits IH cell apoptosis through hsa-miR-138-5p/RGS5 axis.

circular RnA (circRnA) is endogenous non-coding RnA (ncRnA) with a covalently closed circular structure. it is mainly generated through RnA alternative splicing or back-splicing. circRnA is known in the majority of eukaryotes and very stable. However, knowledge of the circRnA involved in regulating cashmere fineness is limited. Skin samples were collected from Liaoning cashmere goats (LCG) and Inner Mongolia cashmere goats (MCG) during the anagen period. For differentially expressed circRNAs, RNA sequencing was performed, and the analysis led to an identification of 17 up-regulated circRNAs and 15 down-regulated circRNAs in LCG compared with MCG skin samples. In order to find the differentially expressed circRNAs in LCG, we carried out qPCRs on 10 candidate circRNAs in coarse type skin of LCG (CT-LCG) and fine type skin of LCG (FT-LCG). Four circRNAs: ciRNA128, circRNA6854, circRNA4154 and circRNA3620 were confirmed to be significantly differential expression in LCG. Also, a regulatory network of circRNAs-miRNAs was bioinformatically deduced and may help to understand molecular mechanisms of potential circRnA involvement in regulating cashmere fineness.

The emerging role of circular RNAs in common solid malignant tumors in children

Malignant tumors are one of the fatal diseases that threaten children’s physical and mental health and affect their development. Research has shown that the occurrence and development of malignant tumors are associated with the abnormal expression and regulation of genes. Circular RNAs (circRNAs) are noncoding RNAs that have a closed circular structure, with a relatively stable expression, and do not undergo exonuclease-mediated degradation readily. Recent studies have shown that circRNA plays an important role in the occurrence, metastasis, and invasion of solid malignant tumors (SMTs) in children. Thus, circRNA is being considered as a breakthrough in the treatment of SMTs in children. In this review, we describe the functions and mechanisms of circRNAs involved in SMTs in children oncogenesis, and summarize the roles of circRNAs in regulating cell proliferation, cell apoptotic death, the cell cycle, cell migrative and invasive ability, epithelial-mesenchymal transition (EMT), cancer stem cells and drug resistance in SMTs in children. In addition, we also discuss the role of circRNAs in the early diagnosis, pathological grading, targeted therapy, and prognosis evaluation of common SMTs in children. CircRNAs are likely to provide a novel direction in therapy in SMTs of children.
