Expression of X-Linked Inhibitor of Apoptosis Protein (XIAP) in Breast Cancer Is Associated with Shorter Survival and Resistance to Chemotherapy

Abstract: XIAP, the most potent inhibitor of cell death pathways, is linked to chemotherapy resistance and tumor aggressiveness. Currently, multiple XIAP-targeting agents are in clinical trials. However, the characterization of XIAP expression in relation to clinicopathological variables in large clinical series of breast cancer is lacking. We retrospectively analyzed non-metastatic, non-inflammatory, primary, invasive breast cancer samples for XIAP mRNA (n = 2341) and protein (n = 367) expression. XIAP expression was a… Show more

“…239 Additionally, XIAP overexpression has also been reported in breast cancer, renal cancer, colon cancer and leukaemia and correlated with poor overall survival, thus, positioning it as a potential therapeutic for cancer treatment. [240][241][242][243][244] In a related study, overexpression of TRIM9 is opined to correlate with the upregulation of Bcl-2 and downregulation of caspases 9/7 leading to apoptosis inhibition in lung cancer. 245 Similarly, Yang et al 246 reported a correlation between TRIM9 overexpression and decreased caspase 3 activity in uterine leiomyoma.…”

“…XIAP, strongly inhibited by CM treatment (−149.92 ± 93.7 fold), is an apoptosis inhibitor; it suppresses apoptosis by binding caspase and inhibiting caspase activation [50]. XIAP modulation is becoming an important target for cancer therapy, in fact, it may confer therapeutic resistance and may modulate signaling factors involved in other processes, such as necroptosis, autophagy and immunosuppression [51]. NOL3 is an apoptosis suppressor and is related to chemoresistance and radiotolerance in cancer cell [52].…”

“…Copper-containing ferroxidase -ceruloplasmin, a large amount of which is concentrated in Sertoli cells, participates in transport of iron ions, which are the basis for such fundamental cellular processes as oxidative phosphorylation and oxygen transport (Vashchenko & MacGillivray, 2013;Tvrda et al, 2015). Other than enzyme cofactor, copper takes part in realization of signal pathways of apoptosis development by inhibiting activation of caspases 3, 7 and 9 (Mufti et al, 2006;Devi et al, 2021). This confirms the leading role of copper in seminal plasma in the control of the number of live spermatozoids.…”
