Expression of the alpha subunit of 7S nerve growth factor in the mouse submandibular gland

Abstract: alpha-NGF is an inactive serine protease that is associated in the mouse submandibular gland with a closely related serine protease, gamma-NGF, and the neurotrophic factor, beta-NGF. The heterogeneity of purified alpha-NGF has been examined by DEAE-cellulose chromatography and SDS polyacrylamide gel electrophoresis. A possible explanation for the observed heterogeneity is presented. Antibodies have been prepared against alpha-NGF and purified by affinity chromatography so that they do not cross-react with gamm… Show more

“…added along with anti-a subunit IgG blocked precipitation of all four of these polypeptides (Figure 4c), demonstrating that immunoprecipitation was specific for all of them. The two higher molecular weight bands consistently appeared as a doublet with molecular weights -between 30,000 and 34,000, consistent with the size previously reported for the a subunit precursor (12,14). The lowest molecular weight band is the same size as the mature a subunit large chain (16,500).…”

“…(Figure 2). The size of the RNA (approximately 1000 bp) was sufficient to code for an oc subunit precursor, which has a molecular weight of approximately 32,000 (12,14).…”

“…It is thus intriguing to hypothesize a cleavage of the MSP precursor by NGF-␥ and epidermal growth factor-binding protein. These could be transported from the submandibular glands, where they are highly expressed (Drinkwater et al, 1987;Isackson et al, 1987), to the brainstem through the extensive connections that link together masticatory, facial, and lingual neuromuscular systems (Fay and Norgren, 1997).…”
