Expression of the Adhesion Molecules CD49d and CD49e on G-CSF-Mobilized CD34+Cells of Patients with Solid Tumors or Non-Hodgkin's and Hodgkin's Lymphoma and of Healthy Donors Is Inversely Correlated with the Amount of Mobilized CD34+Cells

Abstract: The yield of CD34+ PBPC and colony-forming units-granulocyte-macrophage (CFU-GM) in leukapheresis products and the expression of the adhesion molecules CD11a, CD31, CD49d, CD49e, CD54, CD58, CD62L, c-kit (CD117), Thy-1 (CD90), CD33, CD38, and HLA-DR on CD34+ PBPC were analyzed in patients with cancer of the testis (n = 10), breast cancer (n = 10), Hodgkin's disease (n = 20), high-grade (n = 20) and low-grade (n = 20) non-Hodgkin's lymphoma, and healthy donors (n = 20) undergoing G-CSF (filgrastim)-stimulated P… Show more

“…However, in our study the "good mobilizer" group of patients had a higher percentage of cells coexpressing VLA-4 on CD34 + cells compared to the "poor mobilizer" group of patients. In this respect, Zeller et al [49] [2,10,42,44]. The differences between the Zeller et al results and our results could be disease-related and not directly associated with expression of adhesion molecules.…”

“…A few prior studies have also attempted to identify differences in adhesion molecule profiles between good and poor mobilizing patients with conflicting results. Thus, Gazitt and coworkers 4 found a direct correlation between CD49d (VLA‐4) expression and mobilization efficiency whereas Zeller and coworkers 5 showed an inverse correlation with CD49d (VLA‐4) and CD49e (VLA‐5). These differences might suggest that CD49d (VLA‐4) and CD49e (VLA‐5) are, at best, borderline predictive factors, as suggested from our results.…”

“…A number of studies over the past decade have suggested that adhesion molecule expression is important in the trafficking of CD34+ progenitor cells 3 . Only limited and conflicting data, however, have been published comparing adhesion molecule expression profiles on the mobilized cells of good and poor mobilizing patients 4,5 . In this study we compared the expression of eight adhesion molecules by the collected CD34+ cells from 13 “good” and 23 “poor” mobilizing patients.…”

“…Furthermore, Zeller et al . [80] reported an inverse correlation between the amount of CD34 + cells mobilized and the extent of coexpression of CD34 + VLA‐4 + cells. Others also reported a decrease of VLA‐4 expression in mobilized peripheral blood [72, 7480].…”
