Expression of steroidogenic factor 1 and Wilms' tumour 1 during early human gonadal development and sex determination

Abstract: The transcription factors SF-1 and WT1 play pivotal roles in mammalian gonadal development and sexual differentiation. In human embryos, both SF-1 and WT1 are expressed when the indifferent gonadal ridge first forms at 32 days post-ovulation. As the sex cords develop - providing morphological evidence of testis differentiation - SF-1 localises predominantly to developing Sertoli cells in the sex cords, whereas WT1 retains a broader pattern of expression. Later, SF-1 localises predominantly to steroidogenic Ley… Show more

“…In humans, WT1 expression first appears at 32 days postovulation (d.p.o. ) in the gonadal ridges of both male and female embryos and then increases at 41 d.p.o., whereas SRY is most strongly expressed at 44 d.p.o., and both genes are expressed later in fetal development (Hanley et al, 1999(Hanley et al, , 2000. Furthermore, mutations in both genes are associated with gonadal dysgenesis in mice and humans as described above.…”

“…Dated mating was identified by the presence of a vaginal mucous plug which was considered to be embryonic day zero. Mouse embryos of E9n5-13n5 were obtained, fixed in 4 % paraformaldehyde in phosphate buffered saline and embedded in paraffin wax (Hanley et al 1999).…”

“…In developing embryos, Nr5a1 is expressed in the urogenital ridge, representing the first marker of gonadal and adrenal differentiation (Ikeda et al, 1994; Morohashi et al, 1995, 1999; Ramayya et al, 1997; Hanley et al, 1999, 2001). Throughout human development, NR5A1 is expressed in the steroid‐secreting adrenal cortex, in Leydig and Sertoli cells, as well as in granulosa and theca cells.…”
